Liver injury is common a pathological change to many liver diseases, its persistence usually is a important initiating factor for the cause of liver fibrosis, even cirrhosis and liver cancer. Therefore, prevention of liver injury is one of the main link to liver disease prevention in clinical. Complex mechanism of liver injury, with the development of modern medicine, knowledge of liver injury has entered the cell at the molecular level, prevention and treatment of TCM knowledge of the mechanisms of liver injury is also from the protection of the liver cell membrane, reducing transaminase levels gradually shallow depth. Recent studies show that compared with other mechanisms in terms of, apoptosis in the liver cell injury play a more important role. The current mechanism of apoptosis of liver injury which is an important research content, while Chinese medicine research in this direction is more scattered, not forming system.The action of Salvia, Red Peony Root and SanQi powder in ShuGanQingDuTang Bupleurum is sparse liver stagnation, blood conditioning and cooling blood. The action of peony is supporting and softening Liver. Diffusa, Abri, Clover Isodon have effect of heat-clearing and detoxification; which clover Isodon have action of heat-clearing, detoxifying and curing dampness jaundice. Abrus canclear heat and detoxify, Traditional analgesic effect, is commonly used in traditional Chinese medicine clinical treatment of hepatitis. The liver function with various multi-drug, anti-virus, anti-inflammatory effect, Radix, clover tea dishes with regulating the immune function and promote liver regeneration, hepatic fibrosis; Salvia, red peony, SanQi powder also has to improve the hepatic microcirculation, remove immune complexes, the role of inhibition of fibrosis. According to the tendency of the liver injury mechanisms in TCM, based on ConA, BCG plus LPS, D-galactosamine induced liver injury model, we evaluated the action of ShuGanQingDuTang for hepatic injury, and regulated by signaling pathway-related Detection of genes and their expression, to explore ShuGanQingDuTang on liver injury in liver cell apoptosis signaling pathways regulating mechanism.1. ConA induced liver injury:Animals were randomly divided into 6 groups, in addition to the normal control group, the remaining mice after intravenous injection of ConA20mg/kg were on the first day, the afternoon and the next day administered orally once each afternoon. After the last administration 4-6h in model group and treatment group animals single intravenous injection of ConA20mg/kg.12h, eyeball blood, animals were decapitated, and the separation of serum measured SOD, MDA, GSH and NO, for the liver biopsy specimens, and photographs. The results showed that: compared with the control group, model control group ALT was significantly higher (P<0.01), shows the model was successful. DDB, large dose of ShuGanQingDuTang, medium, small doses are on the ALT tended to decrease, especially in the doses most significant (P<0.01). ShuGanQingDuTang mouse IFN-γand TNF-αlevels were significantly lower than the model group, the difference was significant (P<0.01).2. BCG+LPS induced liver injury:The BCG+LPS induced liver injury in mice as animal models, not modeling the control group, intravenous injection of normal saline. Each group were administered 1 day,10 days were administered. Serum ALT and AST levels were measured in liver tissue SOD, MDA, GSH, NO levels. The results showed:The levels of serum ALT, AST were very significant increase, compared with the control group (P <0.01). ShuGanQingDuTang in high dose group as well as DDB, can lower the ALT and AST. ShuGanQingDuTang each dose group were significantly higher in immunological liver injury has reduced the SOD content. ShuGanQingDuTang each dose group and the DDB group can significantly reduce the already elevated liver injury in mice liver tissue MDA content. DDB Group and the drug Shugan Tang large, medium, small doses could reduce the hepatic NO levels (P<0.05).3. D-GalN induced liver injury in rats:The NIH 60 mice were divided into 6 groups, drug treatment for 7 days,6 days, model group and treatment group by intraperitoneal injection of 0.5g/kg D-galactosamine, the establishment of D-galactosamine-induced liver injury in animal models. Through testing liver injury ALT, AST, SOD, MDA, NO, Fas and FasL mRNA gene expression and other indicators of liver histopathology in mice, we have the results showed:drug ShuGanQingDuTang on D-galactosamine induced acute liver injury model, has significant protective effects, can significantly enhance the SOD energy, reduce MDA levels, liver pathology and ease. In addition, ShuGanQingDuTang also reduce NO levels, reduced the expression of Fas and FasL.4. On immune system regulation of:(1) on mouse peritoneal macrophage function results:model group, the phagocytic index and phagocytic coefficient and the normal control group (P<0.01), shows the modeling success. ShuGanQingDuTang on cellular immunity in mice with different degrees of inhibition against the role of immunocompromised mice can phagocytic index and phagocytic coefficient is restored. (2) circulating on the chicken red blood cell-induced antibody levels showed: cyclophosphamide can significantly reduce serum hemolysin antibody production capacity (P<0.01), shows the model was successful. ShuGanQingDuTang of large doses of cyclophosphamide-induced inhibition of humoral immunity in mice showed enhanced trend, the other two weaker dose response; levamisole more significant role (P<0.01).This research applicated ShuGanQingDuTang on various reasons drug-induced liver injury has significant protective effect, and the results showed ShuGanQingDuTang can significantly reducing enzyme, antioxidant free radicals and inhibiting lipid peroxidation and regulation of immune function in the anti liver injury. And by reducing NO, increases Bcl-2/Bax, inhibition of intracellular pathways, reducing the expression of Fas and FasL, the apoptosis pathway inhibition of extracellular signal, reducing apoptosis, thus playing its protective liver injury and inhibit liver cell apoptosis effect. This is the future of Chinese medicine for the prevention and treatment of liver injury to provide new ideas for further development and research to lay the initial foundation of traditional prescription.
