objective: Cardiac muscle protein is the basic of heart's structure and function, myocardial structure and function can be changed due to training of long-term system, This will inevitably lead to the changes of cardiac protein expression of quantity and quality. This study investigate the differential expression and the law of right ventricular muscle protein during the moderate and small intensity aerobic exercise conditions from the sight of quantitative and qualitative and preliminary seeking out the target protein that play an important role in cardiac remodelingin under the condition of exercise stress. Provide experimental evidence for study the exercise intervention of National Constitution and Health, and for exercise rehabilitation of chronic cardiovascular disease.Methods:20 rats were divided into exercise and rest group in random. The exercise group was set up to do treadmill training with medium and small intensity exercise and then killed to extract the total protein of right ventricular muscle for two dimensional gel electrophoresis separation. The spots in differential expression volume more than 5 times are selected as alternative spots for mass spectra (MS) identification. The gels maps were analyzed with the Bio PD quest image analysis software to choose variable proteins and the chosen proteins were identified by ULGRAFL-FLEX-TOF/TOF, detect the gene mRNA transcription of target protein With RT-PCR.Results:After 8 weeks' training, the index of heart weight of exercise group increased 32% compared with that of the control group.In electrophoresis map, the average number of detected protein spot in rest group and exercise group are respectively 530±14 and 547±18. There are 58 changed after exercise:33 increased expression≥2times; 25 decreased expression≥2 times. Protein spots of differential expressed≥5 times were selected as object proteinum in MS identification and 11 protein spots of which were assess, include that Annexin A6 (up5.4),Pyruvate dehydrogenase E1 alpha 1 (up5.3),Aconitate hydratase, mitochondrial (down11.3),Actin, alpha cardiac muscle 1 (up9.5),tropomodulin 4 (up5.5),Long-chain specific acyl-CoA dehydrogenase, mitochondrial (down16.7),Electron transfer flavoprotein subunit bet (down16.1),Methylcrotonoyl-CoA carboxylase beta chain, mitochondrial (down 14.6),Hypoxanthine-guanine phosphoribosyltransferase (down9.7),phosphohistidine phosphatase (up9.6),and 29kDa protein(up5.3). Detection results of RT-PCR show: Annexin A6,Actin, alpha cardiac muscle 1,phosphohistidine phosphatase,Long-chain specific acyl-CoA dehydrogenase,Hypoxanthine-guanine phosphoribosyltransferase,Electron transfer flavoprotein subunit bet mRNA was decline; Pyruvate dehydrogenase E1 alpha 1,Actin, alpha cardiac muscle 1 mRNA was increase. Conclusion:①The proteome of the right ventricular muscle had a notable differential expression change after the medium and small intensity training.②Energy metabolism and Ca2+ regulation level has change and Contractility was enhancement during the moderate and small intensity aerobic exercise conditions, different processes of energy metabolism and calcium ions adjustment have different effects on the movement.③Protein expression is not exactly consistent with the gene expression, show that the influence of exercise to protein express is concerned with the transcription of upstream gene or the translation,decorate of downstream.
