Introduction:By use of the new theory and the technique of proteomics, we aim to delve the differential expression and mechanism of the right-ventricular muscle proteome which is of biology meaning during the process of forming the athletic heart,hope to understand the mechanism medium-intensity aerobic exercise-induced change in the heart,aim to provide a reference to study the recommended intervention of students and National Health,and to provide a basis for chronic cardiovascular exercise rehabilitation.Methods:Fifty-four male SD rats were chosen as the research subject and divided into six groups:the control group(3 groups) and the exercise group(3 groups) according to their athletic abilities and weights.The exercise group was made to take on aerobics exercise(medium intensive: 70%-80%Vo2 max).After 4,8,12 weeks,the rats in both of the exercise group and the control group were weighted,anesthetized to death;their hearts were weighted,and the whole proteins of the right-ventricular muscle in the rats were extracted,after that,the proteins were separated by two-dimensional gels electroghoresis,stained by Coomassie brilliant blue dye.The gels maps were analyzed with the Bio PD quest image analysis software to choose variable proteins and the chosen proteins were identified by ULGRAFL-FLEX-TOF/TOF.Results:After 4,8,12 weeks' training,the heart weight of the exercise group increased 1.7%,3.7%and 3.8%respectively compared with that of the control group,and the index of heart weight was 18.5%,34.5%and 36.6%respectively higher than that of the control group.The difference was significant in statistics.It indicated aerobic training had aroused cardiac remodeling and the heart had changed morphologically.Good 2-DE gels were obtained from this experiment with high matching rate and high resolution.Analyzed with Bio-rad PD quest,the protein spots over 10 times in the whole 267 spots which were differentially expressed after training reached the amount of 55.And 11 spots expressed over 10 times after 4 weeks'training(including a disappeared protein,3 up-regulated,7 down-regulated),so did 13 spots after 8 weeks' (including 2 up-regulated,11 down-regulated),and another 31 spots had the similar expression after 12 weeks'(including 7 up-regulated,24 down-regulated).Four significant proteins from the differentially expressed spots were chosen to identify by ULGRAFL-FLEX-TOF/TOF, they were:Pyruvate dehydrogenase E1 alpha 1(disappeared after 4 weeks' training and down-regulated after 12 weeks' training),SBP1 (up-regulated after 4 weeks'training),SPA1(up-regulated after 4 weeks' training),SCAF1(down-regulated after 4 weeks'training).These proteins were closely related to energy metabolism or the self-protective mechanism,and played an important role during the cardiac remodeling.Inclusion:â‘ After 12 weeks training,the rat heart of the test group became larger.â‘¡after the medium intensity training,the proteome of the right ventricular muscle in test group rats had a notable differential expression change after 4 and 8 weeks' training,and after 8 weeks' training,the proteome had an adaptable change.â‘¢we succeeded in filtrating the objective proteins:Pyruvate dehydrogenase E1 alpha 1, SBP1,SPA1,SCAF1,and the other proteins:Cx43,ATP synthase subunit alpha and CAT and so on.The change of those proteins' expression means the medium intensity training can induce the training rat hearts had a well adaptive changes.It could improve the level of cardiac muscle energy metabolism,strengthen the ability of cardiac muscle stress reaction and adaptation,and meliorate the blood pressure adjustment abilities.
