ObjectiveTo observe the influence of interactions between fetal lung fibroblasts (HFL) and lung cancer cells on matrix metalloproteinase-1,2,9(MMP-1,2,9) expression in three-dimensional co-culture.MethodsGLC-82 and HFL were cultured individually in three-dimensional collagen gels, and GLC-82 and HFL were then co-cultured according to the ratio of 5:1 in three-dimensional collagen gels. After 48 hours, harvested the supernatant, MMP-1 was detected by Western blot and MMP-2,9 were detected by gelatin zymography.ResultObservation of the cells growing in three-dimensional co-culture:In three-dimensional co-culture, HFL and GLC-82 were both growing in gel-culture. And they didn't proliferate,and all of the the cells were growing in singles.For example,the HFL cells were growing in gel-culture like trabs,and the GLC-82 were growing in gel-culture with aggressive forms.The expression of Pro-mmp-2,MMP-2,MMP-9 were detected by gelatin zymography:Semi-quantitative Detection of the MMP-2,MMP-9 expression leves.The GLC-82 groups and the HFL groups hardly expressed MMP-9,but the expression level of MMP-9 in co-culture groups was higher than that in single groups.In co-culture groups,the expression of proMMP-2 was higher than that in the GLC-82 groups (P =0.000<0.05) and the HFL groups (P=0.001<0.05). The expression of proMMP-2 in the HFL cells groups was higher than that in the GLC-82 groups (P=0.000<0.05), The GLC-82 groups didn't express (or only a little) MMP-2,but the expression of MMP-2 in co-culture groups was more higher than that in the GLC-82 groups (P=0.000<0.05) and the HFL cells groups (P=0.000<0.05)Using Western blot assy to detect the expression of MMP-1:The semi-quantitative comparison of MMP-1 expression by using the ratio of IOD (MMP-1) and IOD (β-actin). The expression of MMP-1 in co-culture groups was more higher than that in GLC-82 groups (P=0.000 <0.05) and HFL cells groups (P=0.000<0.05)ConclusionThe interactions between HFL and lung cancer cells may enhance the invasion and metastasis of lung cancer through up-regulating the secretion and activation of MMP-1,MMP-2,MMP-9.
