Lung cancer has been the lending cause of cancer incidence and death up to now. Invasion and metastasis is the main cause of death in patients with lung cancer. The invasion and metastasis of lung cancer are closely correlated with many{actors, such as matrix metalloproteinase(MMP), vascular endothelial growth factor(VEGF).Matrix metalloproteinase(MMPs) are a family of zinc-dependent endopeptidases that have been shown to contribute to tumor progression and metastasis. Both tumor and host-derived MMPs contribute to multiple stages of tumor progression through their ability to. degrade the basement membrane and components of the extracellularmatrix. The association of MMP-2 with malignancy may be related to their ability to degrade basement membrane collagen IV to angiogenesis.Vascular endothelial growth factor is a very important positive regulator of angiogenesis. The study found a variety of tumor cells expressed VEGF protein, such as prostate cancer, lung cancer, stomach cancer, breast cancer,et.Deeply studying the expressions and functions of MMP, VEGF in lung cancer which prompts the mechanism of lung cancer invasion and metastasis can provide an important reference to evaluate the invasion,metastasis and prognosis of lung cancer.ObjectiveTo evaluate the stimulus of VEGF on MMP-2 expression of H460 cells in three-dimensional culture collagen gels.MethodsH460 cells were cultured in three-dimensional culture collagen gels.Give different does of VEGF in different group.The expression of MMP-2 were detected by gelatin zymography.Results(1) H460 cells grow well in three-dimensional culture collagen gels.(2) The expression of MMP-2 is different in different VEGF concentration and at different time.24h after three-dimensional culture collagen gels, taking Sample of each group to run gelatinase zymography, MMP-2 zymogen and live enzymes with higher concentrations of VEGF have different degrees of enhanced;48h later, VEGF at high concentrations, MMP-2 zymogen and activating enzyme activity was enhanced, and the expression of MMP-2 has been enhanced over 24h.Conclusion(1) These results demonstrate that the activity and expression of proMMP-2 and MMP-2 were enhanced by VEDF,and the stimulus is does-time dependent; The higher the concentration of VEGF, the stronger the expression of MMP-2 and its activating enzyme; the longer, the stronger the expression of MMP-2 and its activating enzyme。(2) the three-dimensional culture collagen gels provided a reliable research tool for studying the stimulus of MMPs in the invasion and metastasis of cancer.
