Objective Isolating lactobacillus from healthy female genital tracts,for exploring probiotics to research compound preparation.And explored a variety of therapeutic treatment of female reproductive system diseases theoretical basis for doing the preparatory work to use recombinant lactic acid bacteria.Methods Using traditional morphology analysis to identify lactobacillus,Where a gram-positive, catalase test was negative, rod-shaped morphology of the strains could be initially identified as Lactobacillus. And ideniified as the members of the genus Lactobacillus by growth temperature (15℃,45℃), motility test, nitrate reduction test, gelatin liquefaction test, indole (indole) test, hydrogen sulfide tests. Carbohydrate fermentation test for species identification of Lactobacillus. Screening for hydrogen peroxide-producing lactobacillus.Applying 16S rRNA sequence analysis molecular biology methods to identify it exactly. And then examing predominant strian by performance test. These included:growth experiments, different incubation time changes in broth pH, temperature sensitivity of strains testing, acid-resistant strains of the initial screening of Lactobacillus in vitro antagonism tests, antibiotic sensitivity experiments, the thermal stability test metabolites. In addition, detecting strains of the advantages of plasmid.Results There were 11 strains of lactobacillus isolated and identified from 18 health examination women genital tract,the detection rate was 61.1%.5 Lactobacillus species from 11 strains of lactobacillus were identified.4 were Lactobacillus acidlophilus,3 were Lactobacillus gasseri,2 were Lactobacillus jensenii,l was Lactobacillus crispatus and another one was Lactobacillus salivaius. Only one Lactobacillus was hydrogen peroxide-producing.Obtained the predominant strian and Lactobacillus crispatus the highest homology by molecular biology techniques. Conclusion Using traditional biochemical identification and molecular biology methods to identify lactobacillus. Through predominant strain of the preliminary test, which was a strong acid-resistant and hot-resistant one,and was bacteriostasis in vitro testing. In addition, it did not have plasmid DNA.
