The Anti-Proliferation And Inducing Apoptotic Effects Of Ethanol Extract From Boschniakia Rossica On Hepatic Stellate Cell-T6

Objective:To observe the effects of anti-proliferation and inducing apoptosis by Ethanol Extract from Boschniakia rossica on the Hepatic stellate cell-T6, and provide the experimental evidence for Boschniakia rossica as a new anti-hepatic fibrosis drug.Methods:Hepatic stellate cell-T6 (HSC-T6) was cultured in vitro. The inhibition on the proliferation of HSC-T6 by the ethanolic extract from Boschniakia rossica was measured by using MTT assay. The apoptosis and the ultrastructure of HSC-T6 were observed by fluorescent microscope and transmission electronic microscope, respectively. The cell cycle distribution and the apoptotic rate of HSC-T6 was analyzed by floe cytometry.Results:â‘ MTT assay revealed that the ethanolic extract from Boschniakia rossica could inhibit HSC-T6 proliferation in a dose and time-dependent manner.â‘¡The typical morphological changes of the apoptotic HSC-T6 was observed by fluorescent microscope and transmission electronic microscope:cell surface rnicrovilli disappeared, cell size became smaller, cytoplasm concentrated, nuclear chromatin marginationed, nucleus pyknosis, karyorrhexis, blebbing.â‘¢Flow cytometry analysis showed that the HSC-T6 treated with the ethanolic extract from Boschniakia rossica was arrested in G0/G1 phases. The apoptotic rate increased significantly in drug treated HSC-T6 group compared with the control group and positively correlated with the dose increasement of ethanolic extract from Boschniakia rossica.Conclusions:â‘ The ethanolic extract from Boschniakia rossica had a significant anti-proliferation effect on HSC-T6 in vitro.â‘¡The ethanolic extract from Boschniakia rossica could induce HSC-T6 apoptosis and arrest HSC-T6 in G0/G1 phases.
