Objective:To prepare aloe-emodin nanoliposomes,and to investigatethe effect on gastric cancer cells of aloe-emodin nanoliposome transfectionr-Caspase-3-PDT.Methods:Reverse phase evaporation was used to prepare aloe-emodinnanoliposome. TEM was used to observe morphology of AE-L,nanoparticlesize and zeta potential analyzer Nano-ZS90was uesd to analysis the sizeand potential of AE-L,centrifugation was used to determine encapsulationefficiency,western blot was used to detect AE-L transfected r-Caspase-3gene into gastric cancer cells.After AE-L transfection r-Caspase-3geneinduced-PDT treatment of gastric cancer cells, MTT assay was applied toinvestigate the cytotoxicity,flow cytometry and Hochest33342was used toanalyze the cell apopotosis rate and apopotosis.Results:Produced by reverse-phase evaporation method,themorphological of AE-L was similar to spherical.After AE-L transfection r-Caspase-3,the protein Caspase-3express highly in transfected group. Anenergy-dependent manner was showed in the inhibition effect on humangastric cancer cells by the aloe emodin nanoliposome transfectionr-Caspase-3-induced PDT.AE-L transfection r-Caspase-3mediatedphotodynamic(6.4J/cm2) treatment gastric cancer cells12h later,the gastriccancer cells apopotosis rate increased to37.7%; The karyopyknosis,fragmentation and apoptotic body were also found by Hochest33342.Conclusions:Aloe-emodin nanoliposome can be carrier transfectionr-Caspase-3into gastric cancer cells, the apoptosis of gastric cancer cellswas induced by the AE-L transfection r-Caspase-3-PDT significantly.
