Part I Changes of The Antioxidant Enzymes Activity in Sciatic Nerve and Hippocampus of Diabetic Rats[Objictive] To detect the antioxidant enzymes activity in sciatic nerve and hippocampus of diabetic rats, some of which were intragastric administrated with a-lipoic or resveratrol.[Methods] Fifty SD rats were randomly divided into 5 groups:normal group, diabetic group,α-lipoic group,low-dose of resveratrol group and high-dose of resveratrol group.All rats were intragastric administrated with the corresponding drug or normal saline.Twelve weeks later,rats were sacrificed and sciatic nerves and hippocampus were isolated. The activity of MnSOD and CuZnSOD were detected by using the xanthine oxidase methods, the activity of CAT were detected by ultraviolet spectrophotometry, and the activity of GSH-Px and T-AOC were detected by chemocolorimetry.[Results] Compared with normal group, the activity of MnSOD,CuZnSOD,CAT,GSH-Px and T-AOC in sciatic nerve and hippocampus of diabetic control rats were all decreased(P<0.01).α-lipoic or resveratrol increased the activity of CuZnSOD,GSH-Px and T-AOC(P<0.01), resveratrol increased the activity of MnSOD, which in high-dose of resveratrol group were significant(P<0.01),yet which in a-lipoic group were slightly(P>0.05).[Conclusions] The activity of antioxidant enzymes were decreased; resveratrol could increased the activity of MnSOD;α-lipoic or resveratrol could increase the activity of other antioxidant enzymes. PartⅡEffects of Resveratrol on expression of MnSOD in Diabetic Rats Sciatic Nerve and Hippocampus[Objective] To study the effects of resveratrol on MnSOD of diabetic rats sciatic nerve and hippocampus, and to explore its possible mechanism.[Methods] Fifty SD rats were randomly divided into 5 groups:normal group, diabetic group,α-lipoic group,low-dose of resveratrol group and high-dose of resveratrol group.All rats were intragastric administrated with the corresponding drug or normal saline.Twelve weeks later,rats were sacrificed and sciatic nerves and hippocampus were isolated. The activity of MnSOD and CuZnSOD were detected by using the xanthine oxidase methods, the expression of MnSOD were quantified by using real-time PCR. Sciatic nerve and hippocampus ultrastructure changes were examined in electron microscopy.[Results] Compared with normal group, the activity of MnSOD in sciatic nerve and hippocampus of diabetic control rats were decreased(P<0.01), the expression of MnSOD were increased(P<0.05).α-lipoic did insignificant effect on the activity and expression of MnSOD(P>0.05). Resveratrol increasing the expression and activity of MnSOD(P<0.01). The myelin of sciatic nerves in diabetic group appeared loose, the membrane of Schwann'cells became thin, chromatin marginated, and particle-based. Chromatin in pyramidal cells of hippocampus condensed and marginated, mitochondrial swelled, and myelin loosed.α-lipoic or resveratrol could improve the degenerated changes.[Conclusions] The activity of MnSOD decreased in diabetic rats sciatic nerve and resveratrol increased the expression and activity of MnSOD in sciatic nerves of diabetic rats, which may be benefit to diabetic neuropathy.
