| [Objective]To investigate the mRNA and protein expression of CCNE1 and its splicing variants in gastric carcinoma tissues,and evaluate the relationships between the expression and clinic pathological features,so as to investigate its the relationship with pathogenesis and progression of gastric carcinoma.[Method]Oncomine was used to analysis the expression of CCNE1 in gastric carcinoma.Tumor tissues of 34 patients were obtained from GC patients who had undergone curative surgery at Department of General Surgery of the 1st affiliated hospital, Second Military Medical University, from November 2007 to Jμly 2009. The mRNA expression of CCNE1 and its splicing variants was detected in 34 paired carcinoma and normal tissues by SYBR Green real-time PCR. Meanwhile, tissue chip/tissue microarray (TMA) technique combined with immunohistochemistry method was adopted to detect the protein expression levels of CCNE1 in 34 matched tissues.[Resμlts]The mRNA expression of CCNE1 and its splicing variants in the primary carcinoma tissues was significantly higher than that in normal tissues (P< 0.05), the expression levels of CCNE1 mRNA in stageâ… +â…¡and stageâ…¢+â…£was both highly up-regμlated in gastric carcinoma(P=0.042, P=0.016). We indentified two splicing variants of CCN E1,named CCNE1 typeâ…¡, CCNE1 typeⅢ。The mRNA expression of novel splicing variants in primary cancer were up-regμlated compared to the normal tissues (P=0.015, 0.019).The positive expression of CCNE1 protein was higher in primary carcinoma tissues 41.2%(14/34) than that in adjacent-tumor tissues 0%(0/34) and gastritis tisssues0%(0/34) (P< 0.01). The positive expression of CCNE1 protein might be related to depth of invasion, lymph node metastasis and tumor grades (P< 0.05)[Conclution]Up-regμlated expression of CCNE1 and its splicing variants may contribute to the pathogenesis and progression of gastric carcinoma, and may provide useful information for diagnosis and prognosis of gastric carcinoma. |
