Quartz Crystal Microbalance(QCM)technology is a highly efficient,fast,real-time and in-situ detection technology based on the piezoelectric effect of quartz crystals,which has become more mature and more perfect after decades of research and development since its introduction in the 1960 s,and has a wider range of applications.Not only in chemical physics,but also in biology,medicine and surface science,especially for the detection of large molecules and the process of postgraduate chemical reactions have a wider application prospect.In this paper,based on the detection theory of QCM technology in liquid-phase environment,it is shaped into a biochip based on QCM technology through the selection of quartz crystal,the research design of carrying quartz crystal circuit board,and packaging.Based on the Q-value theory to evaluate the frequency stability performance of biochips in liquid-phase environment,the Q-value greater than or equal to 1000 was determined as the evaluation benchmark,and the biochips with excellent performance were selected for biological experiments.A molecular bond cleavage scanner was selected and evaluated to ensure the smooth execution of the bioassay.Finally,after the selection of the biochips and the molecular bonding scanner,the prothrombin time measurement was used as a pre-experiment,and the prothrombin time was further measured and analyzed based on the feasibility of the pre-experiment.In the prothrombin time measurement,the voltage applied to the biochip was optimized,and the optimal voltage for the liquid phase experiment was determined to be 2 V.The volume of reagents used in the experiment was optimized,and the volume of reagents used in the comparison analysis was found to be more suitable at 20:10.In the analysis of the order of reagent addition,it was found that the TT reagent was added first and then the quality control product was added to give better results.After coating the surface of the biochip with polystyrene film,the analysis showed that the results measured on the surface of the biochip with polystyrene coating were closer to the true values and the standard deviation was smaller.The biochip studied in this paper is simple and miniaturized for the measurement of prothrombin time and thrombin time,and the amount of detection reagents is small,and the results can be measured in about 5 min or even less than 3 min.For each sample,a single analysis is possible,and the measurement can be done after sampling.In addition,it can be optimized and improved to achieve in situ detection,with the advantage of immediate detection,fully reflecting the characteristics of rapid and efficient;another can be improved by the process,the detection of antigens and antibodies,viruses and other applications have some prospects.
