Objective: This research aim is to examine the expression of PCSK9 protein in gastric cancer, analyzed the relationship was with expression of PCSK9 and the clinical significance in gastric carcinoma.Methods: Tissue microarray was used to examine the expression of PCSK9 protein in normal gastric mucosa, atypical hyperplasia, cancer adjacent tissue and gastric cancer tissue. This research collected biopsy with 25 cases of normal gastric mucosa tissue,23 cases of atypical hyperplasia,25 cases of cancer adjacent tissue,52 cases of gastric cancer tissue The expression of PCSK9 was detected in normal gastric tissue, atypical hyperplasia, cancer adjacent tissue and gastric cancer tissue by immunohistochemistry(SP method) and analyzed the relationship between the expression of PCSK9 and clinical significance of gastric cancer. Use of real-time fluorescent quantitative PCR( q RT-PCR) to detect PCSK9 gene in human gastric cancer cells, BGC- 823、MGC803 and MKN- 45 m RNA expression situation.Results: The expression of PCSK9 protein mainly showed in cytoplasm of gastric cancer tissues and brown granules displayed in the cytoplasm of positive tissues. Normal epithelial cells were not stained, we observed staining of basal cells in the normal mucosa tissues. The positive expression rates of PCSK9 protein in normal gastric mucosa tissues, cancer gastric tissues and adjacent tissues were 20%(2/10), 82.22%(37/45), 68.89%(31/45). Respectively compared the expression percentage of PCSK9 in normal gastric mucosa tissues with adjacent tissues of gastric cancer and gastric cancer tissues, the distinction was statistically significant(p<0.05); however, the difference was not statistically significant(p>0.05) in cancer adjacent tissue and gastric cancer. PCSK9 in various pathological changes types of gastric mucosa tissue exist expression. The positive expression rates of PCSK9 protein in atypical hyperplasia, cancer adjacent tissues and gastric cancer tissues were 54.55%(12/23)、58.33%(14/24)、73.07%(38/52),the positive expression rates of normal gastric mucosa was 16%(4/25),Compared normal gastric mucosa tissues with atypical hyperplasia, cancer adjacent tissues and gastric cancer, the difference was statistical significant(p<0.01);Compared atypical hyperplasia, cancer adjacent tissues and gastric cancer tissues, the difference was no statistical significant(p>0.05). The expression of PCSK9 in different pathological types of gastric cancer.The expression rates of PCSK9 in well-differentiated group and middle-low differentiated were 40.00%(4/10)、80.95(34/42).Respectively, the difference was not statistically significant(p<0.05),it means that expression of PCKS9 was negative relationship with pathologic differentiation degree of gastric cancer,and increased expression in gastric cancer;the PCSK9 protein expression has no relationship with age 、infiltration 、gender、TNM staging and depth of invasion(p>0.05),but PCSK9 was close relationship with lymph node metastasis(p<0.01),the difference was statistically significant. The results showed that PCSK9 m RNA in human gastric cancer cells BGC-823、 MGC803、MKN-45 is expressed.This experiment confirmed the PCSK9 have expression in gastric cancer cells.Conclusion: The expression of PCSK9 protein in the gastric carcinoma mucosa tissues was overexpression;The PCSK9 protein expression was negatively correlated to differentiation; PCSK9 has close relationship with gastric cancer lymph node metastases.
