Objective:1. Utilize our laboratory's method to establish heavy dysbacteria mice model based on intestinal predominant flora changes,and then, utilize intestinal predominant flora and the stool of the normal mice separately to re-establish flora;2. Analyze the changes of local and general immunity of the heavy dysbacteria mice, to explore the effect of intestinal dominant flora on immune system.Methods: 1. Mice were dealed by intragastic administration of ceftriaxone sodium, according to the analysis of cecum flora,8g/kg/d doses,8 days to establish heavy-dysbacteria model.2. Utilize 10 species intestinal predominant flora,such as Bifidobacterium and Lactobacillus,dealed by intragastic administration quantitively to re-establish flora;3. Utilize the stool of the normal mice,dealed by intragastic administration quantitively to re-establish flora;4. The changes of secreted IgA andβ-defensin in intestinal mucosa of each group mice were detected by ELISA, mucin2,mucin3 andβ-defensin were detected by RT-PCR method.5.Collect the blood of each group mice for rutine analysis.Results:1. After three weeks of 10 species intestinal predominant flora dealed by intragastic administration quantitively,there were 10 species intestinal predominant flora had recovered,include Bifidobacteriuum Lactobacillus,Bacteroides,Escherichia col,Staphylococcus,Streptococcus,Peptococcus,Veillonella,Enterococcus,Fusobacterium;but the Eubacterium was not detected and Yeasts had not recovered.The re-establish rate is 90.9%。2. After three weeks of the stool of the normal mice dealed by intragastic administration quantitively,there were 9 species intestinal predominant flora had recovered, include Bifidobacterium,Lactobacillus,Bacteroides,Staphylococcus,Streptococcus,Peptococcus,Veillonella,Enterococcus,Fusobacterium;but the Eubacterium was not detected and Escherichia coli and Yeasts had not recovered.The re-establish rate is 81.8%。3. Compared with normal mice, the levels of mucosal SIgA was lower andβ-defensin was higher in heavy-dysbacteria mice.;The levels of mucin2,mucin3,β-defensin were higher of mRNA in dysbacteria mice;After the flora were re-established, the level of mucosal SIgA was step up to turn back,the effect of redominant flora dealed by intragastic administration group was most obvious. The level of P-defensin was step up gradually and the mRNA levels of mucin2,mucin3 were turn back.4. The blood of the mice of intragastic administration of 10 species intestinal predominant flora is the most close to the normal value.Conclusion:1. The methods of intragastic administration of 10 species intestinal predominant flora and the stool of the normal mice in one week all could re-establish flora, and close to normal level in three weeks.The method of intragastic administration of intestinal predominant flora is better than the latter.2. As the re-establishment of flora, the level of SIgA turned back,β-defensin secretion increased, meanwhile, Intestinal epithelial mucin expression showed a recovery trend. Re-establishment of intestinal predominant flora plays an essential role on the promotion of intestinal mucosal immunity.3. The method of intragastic administration of 10 species intestinal predominant flora is more conducive to general immunity of the antibiotic induced heavy dysbacteria mice than the other two methods.
