The D-limonene Impacts On The Expression Of Fascin In Gastric Cancer Cell Line SGC-7901

Objective:To study the molecular mechanism of D-limonene inhibiting the invasion and metastasis of human gastric cancer cell.Method:The RT-PCR and Western Blot methods were used to detect the expression of Fascin mRNA and Fascin protein in SGC-7901 gastric cancer cell that was treated by different concentration of D-limonene(the concentration of D-limonene is 0.0625mmol/L,0.125mmol/L,0.25μmmol/L and control groups.Result:We have detected the expression of Fascin gene in SGC-7901 gastric cancer cell.The result of RT-PCR is that the scalar of Fascin mRNA expression in different groups is insignificant and the size of Fascin mRNA bands is fully consistent with the designation.The time of treatment with D-limonene is 24h.the GAPDH is used as reference substance. We also have detected the expression of Fascin protein in SGC-7901 gastric cancer cell by using Western Blot method.The scalar of Fascin protein is varied with the change of the concentration of D-limonene.The higher of the concentration of D-limonene is,the smaller the expression of Fascin protein is.when the concentration of D-limonene is 0.25mmol/L,the expression of Fascin protein is smallest in the four groups.This is consistent with the experiment in vitro in the pastConclusion:D-limonene can inhibit the expression of Fascin protein. Thereby suppress the invasion and metastasis of SGC-7901 gastric cancer cell.
