Objective:1. To investigate (1) the effects of subchronicl low concentration arsenic on the kidney function and.(2) the early biomarker of the kidney in animals exposure to low concentration arsenic by proteomic analysis.Methods:1.Twelve healthy male rabbits were randomly divided into four groups.They were exposed to different doses of sodium arsenite(0,10,50 and 250μg/L) for 12 weeks through drinking water. Body weights were measured at 0w,4w,8w and 12w.The kidney function(Cr and BUN) were measured at 8 weeks and 12 weeks by colorimetry. Arsenic levels in blood,liver and kidney were detected by Flame Atomic Absorption Spectrometry. The morphological changes of kidney were observed by light microscopy. 2.48 rats were randomly divided into four groups, each of which was treated with arsenic exposure at 0μg/L 50μg/L 150μg/L 450μg/L in drinking water. After 4 week's exposure the rats were killed and the kidneys were collceted for proteomic analysis. The experiment is designed to research the proteomic differences between different doses induced by low arsenic and the rats by using two-dimensional gel electrophore-sis (2-DE) and mass spectrometr and to investigate the new protein, which exists in the rats induced by low arsenic and speculate their function.Results:1. With the extension of time, the body weight has an increasing tendency at the four groups, and the 50μg/L group increases obviously, but there are not significant differences in body weight(P>0.05). Compared with control group, arsenic levels in kidney in 50 and 250μg/L doses showed dose-response relationship (P<0.05);2. Compared with control group, Cr levels elevated as the dose increased. BUN did not change significantly, there are not significant differences in body weight(P>0.05).3. Under light microscope:The nephron in control group constitutes by the cortex and medulla. In cortex, the structure of renal glomerulus is complete and the tubulus have a certain distribution and direction. None of degeneration and necrosis, inflammatory cell infiltration and cell detachment are in tubulus; In low dose group,a large number of inflammatory cells infiltrate into the renal interstitium; Medulla can see a high degree of congestion. The part of lumens of renal tubulus are expansion and epithelium is flat. The epithelial cells have mild abscission. A small number of tubulus emerge from protein.4.2-DE Results:Compared with control group, the protein expression of each group have different levels of increased and lower, there are 4 new protein spots,12 disapperar spots and 21 spots expression increased or decreased appeared in arsenic groups. It is necessary to do a further mass spectrometry to determine the protein's type and quantities.Conclusions:1. The 50μg/L group rabbits has an obviously increasing tendency in the body weight.2. Arsenic may accumulated in the kidney of rabbit exposed to 50 and 250μg/L groups and cause the damage of rabbit kidney function.3. The 50μg/L group appears a large number of inflammatory cells,the 150μg/L group can cause tubules lumens expansion,the epithelial cell abscission and protein tubulus.4. The proteins of 6702,6703,2502,6504,2602,1501,6701,7502,3201,9101, and 9107 may related to the arsenic toxicity of kidney.
