Objective:To study the possible effect of Genistein on cultured human pancreatic cancer cell line BXPC-3 growth inhibition and apoptosis in vitro.Methods:The pancreatic cancer cell group BXPC-3 was established by way of cell culture in vitro. The effect of Genistein on the proliferation of BXPC-3 cells was examined by MTT method. The changes of the cell cycle and apoptosis of BXPC-3 cells dealing with Genistein were determined by flow cytometry.Results:The MTT results showed that the effect of different concentrations of Genistein on BXPC-3 cells has a statistical significance between the experimental group and the control group after 24h and 48h (P<0.01).The inhibition rate of high concentrations group was higher than the low concentration group and the inhibition rate in 48h was higher than in 24h.Flow cytometry analysis showed that the cell cycle could be changed by Genistein.G2/M phase cells became increased and S-phase cells,G0/G1 phase cells reduced with the increasing concentration and the role of extended of Genistein.Genistein could promote the apoptosis of the BXPC-3 pancreatic cancer cell and with the increasing concentration and time-extended to 48h of Genistein,apoptotic rate was also increased.The difference has a statistical significance between the different concentration of groups (P<0.05).Conclusion:Genistein can inhibit human pancreatic cancer BXPC-3 cell proliferation and induce apoptosis in vitro,both radiate a time-and dose-dependent.
