Carcinoma of the uterine cervix is the second commonest female cancer worldwide.The Prevelance lies in first Place in our country (China) and become one of the main disease which Severly threaten health of women in our country.Lymph node metastasis is a independent Prongostic factor and the main death.cuase of cervical cancer Patients.In order to understand carcinogenesis, development and lymph node metastasis mechanism of cervical cancer,our previous study had selected the differential expression gene bye DNA gene chip and found many genes correlated strongly with lymph node metastasis of early squmaous cervical cancer. In these genes, the expression of MIF in early squmaous cervical cancer with lymph node metastased was higher 3.4 times than with no metastatic lymph node.we speculate MIF Plays an important role in cervical cancer invasion and metastasis.Purpose:To determine the correlation between the expression of (macrophage migration inhibitory factor MIF)and Lymph node metastases of squamous cervical cancer and to elucidate the mechnaism of MIF-mediated invasion nad metastases of earlier squamous cervical cancer and to Provide a Possible new clinical way for the therapy of cervical cancer.Methods:migration of cervical cancer cells (caski,siha,c33a)and angiogenesis by Vitro Angiogenesis Assya Kit. and look for the best stimulus to the next experiment.he expression of MIF Portein and intratumor matrix metallo Proteinase-(MMP-9) and Extracellular signal-regulated kinase (ERK)in squamousee cerviel cancer were decideded respective by antibody againstMIF associated antigen and Immunohistochemieal analysis.The expression of MIF,MMP-9 portein in blood serum was detected by ELISA and western-blot.Recombinant human MIF stimulate eevrical cancer cell(siha and caski and c33a) and Human umbilical Vein endothelial cells(HUVEC) to detect MIF-mediated cervical cancer cells and HUVEC Proliefration and utmo-rnedohtelial adhesion by MTT analysis.Result:The cervical cancer with lymph node metastases express MIF,Extracellular signal-regulated kinase (ERK) and MMP-9/2.MIF at singifienatly higher level than negative metastatic lymph node(=P0.00,0.02).othewrise, the express of MIF coerrlated strongly and Positively with MMP-9/2 of cevrieal cancer(r=0.251, P<0.05).Eetogenie MIF stimulate cevrieal cancer cells and HUVEC Proliefration, migration, enhance Tumor-rendohtelial adhesion and Pormote cpaillary tube fomration.MIF over-expressing cells.Conclusion:MIF mihgt enhance early cervical cancer to invase and metastases by stimulate Dieretly cervical cancer cells Proliefration, migration and enhance tumor-endohtelial adhesion.On the other hand, MIF mihgt Stimulate HUVEC proliefration, migration and promote Palliyar Tube of mrationtoin directly enhance cervical cancer to invase and metastases, theratenedment Targeted MIF mihgt to provide a new clinical way of therapy of cervical cancer.
