Objectives To investigate bacterial translocation and its mechanism in children with intussusception; Methods To detect the presence of bacteria DNA (16SrRNA shared by all bacteria and E.coli BG) in whole blood using polymerase chain reaction (PCR). Bacteria in mesenteric lymph nodes were cultured. Bcl-2 and Bax in intestinal tissue were determined by immunohistochemical assay.Enzyme-linked immunosorbent assay(ELISA)was used to detect the level of IL-8 and IL-11 in serum;Result Bacteria DNA were not detected in the control group; Air reduction group and operation reduction group 16SrRNA positive rates were 30%,50%, BG positive Rates were 20%,60%;The intestine resection group 16SrRNA positive rates were 60%, BG positive rates were 70%;In operation group and the intestine resection group, positive cultural rate of E.coli in mesenteric lymph nodes was 50% and 60%. Intussusception groups tissue Bcl-2 and Bax with an average of gray were different from the control group, statistically significant (P<0.01), it was significant within group (P<0.01).Intussusception pre-operative group, the concentration of IL-8 and IL-11 were different from the after-operative group (P<0.05) Intussusception pre-operative group and the control group were statistically significant (P<0.01),there were difference within appendicitis pre-operative group (P<0.01), the control group was not different from the after-operative group (P>0.05)Conclusion Polymerase chain reaction(PCR) can early detection of bacterial translocation in children with intussusception. The expression of Bcl-2 and Bax were induced by intestinal ischemia reperfusion in intussusception, then intestinal apoptosis and eventually lead to the mechanism of bacterial translocation.Bacterial translocation maybe correlated with IL-8, The serum IL-11 level is of great implication to predict bacterial translocation and progrosis.
