Objective To investigate the expression of HIF-1α,VEGF and Glut1 in gastric cancer (GC) tissues and their relationship with clinical pathological features,to explore the efforts of lacking of oxygen on human gastric cancer cell line BCG-823 to analyse the expression change of HIF-1α,VEGF and Glut1 protein. Through this research, we hope to explore their roles in progression of gastric carcinoma and to evaluate their clinicopathologic significances. Methods Part 1: The expression of HIF-1α,VEGF and Glut1 proteins was detected in GC and adjacent tissues by immunohistochemical staining(IHC), and compared with the clinico-pathological data. Part 2: We treated human low-differentiated gastric adenocarcinoma cell line BGC-823 with or without CoCl2, the efforts of on expression of HIF-1α,VEGF and Glut1 were measured by immunocytochemical staining(ICC) and immunoflurescence assay(IFA). MTT assay was used to evaluate the growth of BGC-823. Results Part 1: Positive rates of expression of HIF-1α,VEGF and Glut1 were 70.0%, 61.7% and 69.1% in GC, and 21.7%,11.7%,18.3% in adjacent tissues, respectively. The expression of all three proteins showed a significant difference between the GCs and adjacent tissues (P<0.05). The positive rate of HIF-1α,VEGF and Glut1 expression was significantly related to the depth of invasion,lymph node metastasis,TNM stage of GC. In addition, HIF-1a,Glut1 expression correlated with histopathologic type ,VEGF expression correlated with cell differentiation(P<0.05). The positive expression of HIF-1a were positively correlated with VEGF and Glut1(p<0.001). MVD of the positive expression of HIF-1a is obviously higher than the negative expression of HIF-1a.Part 2 : [1] The expression of HIF-1α,VEGF and Glut1 were measured at different levels without CoCl2. After exposure to CoCl2 (200μmol/L),the expressions of HIF-1α,VEGF and Glut1 are increased.[2] After exposure to CoCl2 (50μmol/L,100μmol/L,200μmol/L ), the growth of GCs was promoted. Meanwhile the inhibitory effect of CoCl2 (400μmol/L,800μmol/L) was observed unexpectedly. Then the cell growth curve was measured at different time (8h,16h,24h). As a result, it showed time-dependent manner. Conclusions [1] The over-expression of HIF-1α,VEGF and Glut1 in GC were relatived with the happening of GC; They are the very important roles in the process of invasion and metastasis of GC; HIF-1αmaybe a very crucial factor of tumor neovascularization and metabolism. [2] The tumor neovascularization and metabolism are relatived with hypoxia microenvironment when the gastric cell is in a certain condition of hypoxia.Hypoxia microenvironment is one of the crucial factors of the gastric cell proliferation.
