Part oneStudy on the expression of HIF-1αand VEGF in QBC939 cell line under chemical hypoxia induced by COCL2Objectives:To examine the expression change of HIF-1αand VEGF in protein levels and the correlation of them in QBC939 cell line under chemical hypoxia induced by COCL2 in vitro.Methods:The establishment of different concentrations of COCL2 groups (200,150,100,50μmol/L),control group without COCL2.The establishment of different periods (6h,12h,24h,and 48h)groups in hypoxia.Different concentrations of COCL2 to interfere cholangiocarcinoma cell QBC939 and150μmol/L COCL2 interfere cholangiocarcinoma cell QBC939 at different periods.ELISA was used to determine the change of VEGF protein in cell culture supematants on different COCL2 doses and periods groups.Immunohistochemical technique was used to determine the change of VEGF and HIF-1αprotein in QBC939 cell.Results:COCL2 can up-regulate the expression of HIF-1αand VEGF at least partly in a dose and time dependent pattern.The levels of VEGF protein in the different COCL2 concentrations groups of cell culture supernatants were higher than control group(P<0.05).The VEGF protein also increased continuously with the growing of hypoxia time(P<0.05).The average grays of HIF-1αand VEGF protein showed a significant difference between any COCL2 concentrations group than control group(P<0.05).Different time periods of hypoxia HIF-1α, VEGF protein in the average gray,Any two group comparison have difference(P<0.05). HIF-1αwas positively correlated with VEGF protein expression in the different COCL2 concentrations groups r=0.830,P<0.01.HIF-1αwas positively correlated with VEGF protein expression in the different periods groups,r=0.909,P<0.01.Conclusion:In QBC939 cell line of hypoxia,there is up-regulation HIF-1αand VEGF. HIF-1αtakes the role of regulation the expression of VEGF. Part twoEffect of somatostain analogue(OCT)on the expression of HIF-1α, VEGF in QBC939 cell line under COCL2-induced hypoxiaObjectives:To evaluate the effect of somatostaln analogue(OCT)on the expression of hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF)in QBC939 cell line under chemical hypoxia induced by COCL2,and to explore its inhibitive on cholangiocarcinoma cell line QBC939.Methods:Cholangiocarcinoma cell line QBC-939 were cultured in vitro under chemical hypoxia induced by COCL2.The establishment of different concentrations of OCT groups(5, 0.5,0.05,0.005mg/L),control group without OCT.Different concentrations of OCT to interfere cholangiocarcinoma cell QBC939.ELISA was used to determine the change of VEGF protein in cell culture supematants on different OCT doses groups.Immunohistochemical technique was used to determine the change of VEGF and HIF-1αprotein in QBC939 cell.Results:The levels of VEGF protein in the different OCT concentrations groups of cell culture supematants were lower than control group(P<0.05).The average grays of HIF-1αand VEGF protein showed a significant difference between any OCT concentrations group than control group(P<0.05).HIF-1αwas positively correlated with VEGF protein expression r=0.973,P<0.01.Conclusion:OCT can inhibit the expression of HIF-1αand VEGF in QBC-939 cell under hypoxia,which may be one of the anti-tumor mechanism,Meanwhile,OCT may be through inhibition of HIF-1αto inhibit VEGF expression.
