Objective:Esophageal cancer was Common digestive tract tumors,which serious threated human health. It happened were related with inherentgene, environment, living habits, and so on. While looking for the methods oftumor treatment, Found that biological rhythm disorders can lead to thehappening of tumors and progress. As material base of biological rhythm,circadian gene per2play a very important role in the progress of tumor. Thisexperiment is in vitro culture of Eca-109cells,to Test the phase of circadianrhythm genes per2and VEGF in dark and light-dark cycle in Eca-109cells.Identification of the best concentration of CoCl2hypoxia induced andhyperplastic toxicity in different concentrations on esophageal cancercells.Hoping to provide some theoretical evidence for chronotherapy.Methods: Using to avoid light low oxygen and low oxygen asexperimental group and control group respectively in vitro cultivate of Eca-109cells. To Test the phase of per2and VEGF by real-time quantitative PCR,CCK-8testing different concentration of CoCl2hypoxia induced the Eca-109cells.Results: No matter in continuous darkness or normal light-dark cycle,theexpression of per2and VEGF were spontaneous rhythmicity,the phase of per2and VEGF were opposite. CoCl2induced the expression of VEGF in Eca-109cells,And Different concentrations have different hyperplastic toxicityinfluence on the Eca-109cells.Conclusion: the Phase of per2and VEGF were changeless, Regardless of under in dark and light-dark cycle. per2in a negative regulatory role onthe expression of VEGF. The hypoxic effect is best in150μmol/Lconcentrations induced by CoCl2,there were obvious proliferation inhibitioneffect under high concentration (250,300,400(including mol/l) for Eca-109cells.
