Fluorescence spectra of active components in traditional Chinese medicine, praeruptorin A and umbelliferone, were studied in this thesis. Influences of experimental conditions on fluorescence spectra were also investigated. The fluorescent components of Peucedanum praeruptorum Dunn (PPD, Baihuaqianhu in Chinese) and Peucedanum Decursivum (PD, Zihuaqianhu in Chinese) were basically separated by column chromatography. Fluorescence spectra of some components were studied. Three-dimensional fluorescence spectra of PPD and PD were primarily analyzed, and the differences between them were explained.The main contents of the thesis were as follows:1 Fluorescence spectra of praeruptorin A and umbelliferone were studied. Influences of solution acidity, concentration and content of methanol on the fluorescence spectra were explored. It was found that solution acidity had great effect on the fluorescence spectra of praeruptorin A and umbelliferone, and umbelliferone produced strong fluorescence in weak alkaline condition. The fluorescence quantum yields of praeruptorin A and umbelliferone were measured to be 0.11 and 0.91 respectively, using quinine sulfate as a reference. The ionization constant of umbelliferone was determined to be pKa=7.61, by fluorescence method.2 Fluorescence components of PPD were basically separated by column chromatography and their fluorescence properties were studied. It was found that fluorescence spectra of an obtained pure component were similar to the fluorescence spectra of praeruptorin A. Three-dimensional fluorescence spectra of PPD were primarily analyzed.3 Fluorescence components of PD were separated by column chromatography and their fluorescence properties were studied. A component with strong fluorescence was studied by mass spectrum, and was estimated to be Nodakenin. Three-dimensional fluorescence spectra of PD were analyzed, and the differences between PPD and PD were discussed.
