Objective : To observe the effects of salvianolate on collateral blood vessel and heart function in acute myocardial infarction (AMI) model in swines.Method : Twenty-one young swines were randomly divided into three groups: control group, high-dose salvianolate-treated (HS) group and low-dose salvianolate-treated (LS) group (n=7, in each group). AMI model was made by ligaturing left anterior descending coronary artery of swines. A dose of 400 or 200mg salvianolate dissolved in 250 ml 5% glucose in normal saline were administered by intravenous drip to swines in the HS group and LS group respectively after operation for 7 days, and the swines in the control group were administered with 250 ml 5% glucose in normal saline synchronized. 4 weeks after operation, coronary bypass circuit and LVEF were evaluated by coronary arteriongraphy, immunohistochemistry and gated myocardial perfusion imaging respectively. And cardiomyocytic apoptosis were detected by TdT-mediated dUTP-biotin nick end labeling (TUNEL) method. Apoptosis index was calculated under an optical microscope.Result :(1) Coronary arteriongraphy: infarction region of therapic group collateral circulation outweigh control group obviously,esespecially in high dose group. (2)Gated myocardial perfusion imaging:LVEF in two therapic groups is higher than control group.(3) Immunohistochemistry: capillary density of treatment group significantly higher than control group,but no significant deviation between high dose and low dose.Similarly, function density of treatment group also significantly higher than control group,and high dose was higher than low dose. (4) Cardiomyocytic apoptotic index in the edge of myocardial infarction was decreased in HS and LS groups, which in HS group was lower than that in control group (P<0.05). (5) Radioactive defect regions were found by gated myocardial perfusion imaging in all the three groups, but that in HS group and LS group were less than those in control group.Conclusion: Salvianolate administered by intravenous drip can inhibit the cardiomyocytic apoptosis, promote blood vessel regeneration, promote formn of collateral blood vessel and improve the function of heart after AMI.
