The present experiment was designed to study the effect of double protein milk for the nomal mice's growth,the preparation of soybean peptide and the effect for the high-fat mice's blood fat and body fat by the double protein milk coordination experiment,the enzymolysis of soybean protein isolate,the hypocholesterolemic activity of soybean peptides in virtro experiment,the foundation of mice's high-fat and the animal experiment.1,Study on technology of the double protein milkThe formula and process technology of double protein with soybean protein and milk protein have been studied, which was made of soy protein isolate, fresh milk, emulsifier, thickener. The experiment have studied process parameters as the content ratios of soy protein solution and fresh milk, the types of emulsifier and addition ratio of compound emulsifier, the types and addition ratio of thickener, homogenization and so on. The best formulas and the processing craft were determined by means of the finished product viscosity, sedimentation rate, pH value, sensory evaluation and other indicators.It was that the mixture ratio of soy protein solution and milk was 2:3, the monoglyceride was 0.2%, the sucrose ester was 0.16%,the carrageenan was 0.005%, the xanthan gum was 0.09%, the CMC was 0.04%, the homogenization was 25MPa, the sterilization was 119℃,10min. The double protein milk was pure flavor, color white, uniform texture and smooth taste, and amino acid score was significantly better than regular milk and soy milk.2,The animal experiments of double protein milk nutrition evaluation30 male healthy Kunming mice were randomly divided into three groups numbered 1-30. A group was fed 12% soy protein hydrolyzate, B group was fed sterilized milk, C group was fed double protein milk of 100ml each day.The result of 7-week feeding experiment proved that,the double protein milk could powerfully increased the weight of experimental rat femur(P<0.05), while experimental rats with increase body weight, blood hemoglobin levels to improve the trend, but the liver weight and liver factor coefficient , heart and heart coefficient, kidney and renal coefficient, spleen and the spleen coefficient, bone length had no significant effect.3,The determination of double-protease of soy protein isolatePlant protease and papain were regarded as the compound enzyme of soy protein isolate (SPI) in comparison with hydrolysis effects of neutral proteinase enzymiatic, alkaline protease, papain, plant protease. The premium working conditions for enzymolysis were determined by means of single factor and L9 (34) orthogonal experiments.The optimal enzymic hydrolysis parameters was that the enzymic hydrolysis temperature and initial pH value was 55℃and 7.0, the substrate concentration was 12%, the quantity of proteinase was 8%, the mass ratio of plant protease to papain was 1:2, hydrolysis rate can reach 14.2%.4,The hypocholesterolemic activity of soybean peptides in virtro experimentThe hypocholesterolemic activity of the hydrolysate of SPI with 8.27 %16.57 %DH which was prepared by two protease was studied.The results indicated that the sample prepared by hydrolyzing SPI with lant protease and papain at DH=14.71% shown highest hypocholesterolemic activity among all the samples, the inhibitory ratio of micellar solubility of cholesterol is 61.67 %.5,The animal experiments of Hypocholesterolemic activity of soybean peptides33 male healthy Kunming mice were randomly divided into three groups. There are 13 mice in group A and fed basic feeding, 7 mice in group B and fed high-fat feeding until the high-fat mice model was founded, the study for soybean peptide solution's lowing fat for high-fat mice was started.; There are 13 mice in group C and fed high-fat feeding.A,C group drink water free and B group was fed soybean peptide solution. Six weeks later , blood serum TC,TG and HDL-C were significantly (p<0.05)by the same way and the the high-fat mice model is succeeded. The lipid-lowering of High-fat mice of 4-week experimental results showed that: the source of cholesterol-lowering soy protein isolate can significantly decrease the concentration of plasma TG and increase mice liver factor (P<0.05). At the same time, the biological active peptides also lower body weight, body weight growth and blood levels of total cholesterol, high density lipoprotein cholesterol levels and low-density lipoprotein cholesterol to some extent.
