GAPSGF is a kind of polysaccharide components of estracting from dry fruiting body of Ganoderm applanatum Pat.The exiting research evidences show that GAPSGF has the efficacy of anti-cancer and the immune system.Therefore,it has the great meaning in theory and practice to extract,separation and purification and preliminary research of component of GAPSGF.It can also apply technical theory for activity,exploitation of new drugs and the quality control of GAPSGF.Objective:To systematically study the GAPSGF, clarify methods of extract,separation and purification to GAPSGF and discuss the component,which will provide scientific basis for researching activity and new drug of polysaccharide.Methods:1. Alcohol precipitation by boiling dried fruit body on the Ganoderma applanatum to extract the GAPSGF.2. In addition to protein Sevage method used, 10% H2O2 oxidative decoloration,dialysis.Purified polysaccharide Ganoderma GF detection by UV spectroscopy,and by DEAE-52 cellulose ion exchange column chromatography.3. The flow of points determined by freezing point lower the molecular weight,and then by TLC.4. By phenol-sulfuric acid method,to measure the content of GAPS GFa,GFb,GFc,GFd.Results:1. Extracted by Ganoderma applanatum,the result show that the extraction rate of 2.126‰.2. In addition to protein by Sevage law, and 10% H2O2 oxidative decoloration,there been purified GAPSGF,and then by DEAE-52 cellulose ion exchange column chromatography,initially four kinds of flow points,respectively GFa,GFb,GFc,GFd.3. Through freezing point depression method, GFa,GFb,GFc,GFd molecular weight were 1159.33g/mol,3654.54 g/mol,3821.19g/mol,1246.85 g/mol.The flow points obtained by TLC, GFa by D-Gal,L-Rha two monosaccharide composition, GFb by D-Glu,D-Ara,D-Gal and L-Rha composed of four monosaccharide sugar,GFc by D-Glu,D-Xyl and D-Ara of three monosaccharide composition,GFd by D-Glu,L-Rha monosaccharide composition of two.4. Through determination on GAPS GFa,GFb,GFc,GFd,their contents were 15.42%,17.25%,18.52%,14.72%,the best hydrolysis time was 6h,5h,5h,6h.Conclusion:1. The extraction rate of GAPSGF was 2.126‰.2. Flow points GFa,GFb,GFc,GFd molecular weight were 1159.33g/mol,3654.54 g/mol,3821.19g/mol,1246.85 g/mol.3.
