| "Shanglu" (Radix Phytolaccae), is the radix of Phytolacca esculenta Van Houtte (P.acinosa Roxb.)or P americana L., as traditional Chinese drug collected in Chinese pharmacopeia, belong to the family of Phytolaccaceae. Shanglu is bitter in flavor and cold in nature. Its parched materials was used mainly in clinical, attributive to the lung, kidney and large intestine meridians, and used for application for treatment of edema, dysuria, antivirus, diminishing inflammation.The physical and chemical identification to distinguish raw and parched Radix Phytolaccae was mentioned in the People's Republic of China Pharmacopoeia (ChP)(2010, Vol(1)), used TLC method to indicate Esculentoside A, and used the content of Esculentoside A(EsA) as indicator, which can not reflect the existence of other compositions.Triterpenoid saponins is the main group of beneficial components responsible for the therapeutic activities and the purpose of heating and vinegar was to destroy some poisons and improve clinical effect, as reported.In our study, Radix Phytolaccae was processed under different temperature and time so that the appearance accord with the criterion of ChP(2010, Vol(l)). The Radix Phytolaccae was extracted on the basis of polarity of triterpenoid saponins so as to establish the TLC and HPLC fingerprints of 1-butanol extracts respectively for Radix Phytolaccae before and after processing, in which the change of components would described.,and the content of Esculentoside A was determined by HPLC-ELSD.The dissertation is divided into three parts, as follows:In the first part, the 1-butanol extracts of Radix Phytolaccae was identified by TLC. The experimental results indicate(?) that the number of spots was same in raw materials of different source, the main diffreence lies in the color of spots. The results indicate(?)that the content of chemical compositions was different. There was no new spots came up after parch process, and the tendency of color changing was also the same in parched Radix Phytolaccae comparing the raw. The results indicate that TLC fingerprints of 1-butanol extracts cannot distinguish raw and parched"Shanglu".In the second part, the HPLC fingerprint of 1-butanol extracts in Radix Phytolaccae was established. Comparing different source and different parched "Shanglu", after processing it was found that the contents of some components were different, seavals change significantly,and found no new component.In the third part, the content of Esculentoside A was determined, different sources and 4 different parched conditions of Radix Phytolaccae were determined.The fingerprints of TLC and HPLC had been established, in which the characteristics of 1-butanol extracts was showed about Radix Phytolaccae before and after processing,the results of the study can reflect the integrity of chemical composition in Radix Phytolaccae more comprehensively. The results suggested the changes of processing conditions affect on the amount of components at different degree. The results of the content detemination showed that Esculentoside A varied significantly in raw and parched Radix Phytolaccae. The content of Esculentoside A increased after processing, the changes depended on parched cotlditons,with steamed time longer and vinegar stewed temperature higher,the content of Esculentoside A rises.Although the appearance was accorded with the cyiterion of ChP(2010, Vol(1)) under different temperature and time, there was no criteria of parched temperature and time described according to ChP(2010, Vol(1)).The difference of parched processing would result in the difference of the pharmacological effects of Radix Phytolaccae.The relationship of the pharmacological effects of Radix Phytolaccae with the influence of parching process need research still. |
