Studies On Chemical Constituents And Bioactivities Of The Flower Of Gentiana Macrophylla Pall.

Radix Gentianae Macrophyllae,one of the well-known traditional Chinese medicine "Qinjiao",is the dried root of Gentiana macrophylla Pall.,which belongs to the Fam.Gentianaceae. Gentiana macrophylla Pall, mainly distributes in Province of Shaanxi and Qinghai.It is embodied as one of the original plants of Radix Gentianae Stramienae in the China Pharmacopeia (2010 edition), which can relieve rheumatic conditions, and remove damp-heat. Wether the flower of Gentiana macrophylla Pall, can be a new medical part is the main purpose of our study. This research introduced the chemical constituents and bioactivity of the flower of Gentiana macrophylla Pall., and it neluded the following three sections.Several isolation methods were used for a preliminary research on chemical components of the flower of Gentiana macrophylla Pall, produced in province Shaanxi. Eleven compounds were achieved finally and they were elucidated by the ways of speetroscopy (UV,IR,MS,13C-NMR,1H-NMR) assisted by comparing with reference substance. They were:β-sitosterol (Ⅰ),daucosterol (Ⅱ),gentiopicroside (Ⅲ). isovitexin (Ⅳ),vitexin (Ⅴ),sweroside (Ⅵ) swertiamarin (Ⅶ),isoorientin (Ⅷ),6'-O-β-D-glucopyranosyl gentiopicroside (Ⅸ),loganic acid (Ⅹ),isoorientin-4'-O-glucoside (Ⅹ)On the basis of having systematically isolated chemical composition of the flower of Gentiana macrophylla Pall.,eight compounds were determined by HPLC-MS: gentiopicroside,vitexin,sweroside,swertiamarin,isoorientin,6'-O-β-D-glucopyranosyl gentiopicroside,loganic acid,isoorientin-4'-O-glucoside. These studies supply basic research project of Pharmacopoeia of People's Republic of China Edition 2010:Quality specification research of Radix Gentianae Macrophyllae. We determined the apparent oil/water partition coefficient of Gentiopicroside by HPLC.We studied the bioactivities of the extract of the flower of Gentiana macrophylla Pall. in the third part. We investigated the effects of anti-inflammatory and anti-analgesic and the effect of gastrointestina motility of mice with constipation. The results showed that the extract of the flower of Gentiana macrophylla Pall, displayed marked and identical anti-inflammatory and analgesic effects. It had obvious effect on constipation model mice caused by diphenoxylate and can enhance the mice's function of gastrointestinal peristalsis. We provided the original research to increase new medicinal parts and expand medicinal plant resources.
