Gentiana macrophylla Pall.is a traditional Chinese medicine in our country.Gentiana exhibits good anti-inflammatory and rheumatoid arthritis effect,and it is used clinically to protect the liver and prevent rheumatism.The literature reports that Gentiana macrophylla is rich in flavonoids and lignans.Recent studies have found that Gentiana macrophylla still contains large amounts of iridoids and phenylpropanoid compounds.In this study,some extracts of Gentiana macrophylla were screened for anti-inflammatory activity.We selected a model about the growth and proliferation of RAW264.7 cells which stimulated by LPS to screen for extracts that exhibit good antioxidant properties.The selected extracts were mainly separated by various chromatograms.The pure compounds was determined by 1H-NMR,13C-NMR,DEPT135,HSQC,HMBC etc.About this research 20 compounds were identified.Compounds structure including:Berchemol-4'-O-?-D-glucoside(1),Phenethanol-?-vicianoside(3),6'-O-?-D-glucopyranosylgentiopicroside(8),Scabrans G3(9),Olivieroside C(7),Methylconiferin(4),Phenolic glucoside(5),Dauco sterol(17),6-(3',4'-dihydroxystyryl)-2-pyrone-4-O-?-D-glucopyranoside(6),?-sitosterol(18),Gentiopicroside(10),Swertiamarin(11),Sweroside(12),Logannic acid(13),(-)-syringaresinol-4,4-bis-?-O-D-glucopyrannoside(2),Blumenol C(14),Byzantionoside B(15),Dibutylphthalate(16),Adenosine(20),P-D-fructofuranosyl-?-D-glucopyranoside(19).Compounds 1,3,4,5,6,7,20 are firstly isolated from Gentiana macrophylla Pall..The experimental data of DPPH radical scavenging rates of iridoids,benzoic acids,and carotenoids showed that all three compounds can eliminate DPPH free radicals in vary degrees;among them,the DPPH clearance rate of sweroside is as high as 80.13%,which excellent oxidation resistance was demonstrated.The data of the reaction rate of the free-radical model with time showed that the scavenging rate of the sample was greatly changed and positively correlated with the concentration of the DPPH solution.This subject provides an important theoretical basis for the research on the antioxidative substances and the classification of medicinal materials of Gentiana macrophylla in the future.It has expanded the composition index research system of Gentiana macrophylla and improved the taxonomic study of Gentiana macrophylla.
