Objective: To learn the relationship between OY-TES-1 gene variants in glioma and OY-TES-1 serum antibody. To explore the reasons of OY-TES-1 antibody positive in glioma patients, to understand wether OY-TES-1 gene is abnormal. Set OY-TES-1 basis for the biological function.Methods: (1) 11 tumor tissue samples from glioma patients with OY-TES-1 antibody-positive. Isolate the total RNA, synthesis cDNA, use reverse transcription-polymerase chain reaction (RT-PCR) technology to amplify OY-TES-1 gene with complete open reading frame sequence. Recover the PCR products and purificate. Undergo the connection of T vector and the extraction plasmid, then do the DNA sequencing, With blast comparison analysis, analysize OY-TES-1 gene sequence situation. Comprehensive analysize the relationship between OY-TES-1 antibody positive and OY-TES-1 gene abnormality.(2) Then to investigate whether OY-TES-1 protein exists in glioma tissues, 6 cases and 4 cases of glioma tissues randomly selected was detected with IHC and WB respectively.Results: (1) OY-TES-1 mRNA can be detected in the 11 glioma patients with OY-TES-1positive antibody; No gene variant was found; in OY-TES-1 gene open reading frame by DNA sequencing.(2) OY-TES-1 protein was mainly localized in the cytoplasm of the tumor cells. Western blot analysis showed that native protein extracted from glioma tissue can combine with OY-TES-1 antibody, but the molecular weight of immunoreactive band was smaller than theoretical value of 61 kDa of OY-TES-1 proteinConclusion: The appearance of OY-TES-1 positive antibody in glioma patients may not be caused by OY-TES-1 gene variant. The specific mechanisms still need to be further studied.
