Objective:To explore the alterations ofα2-Antiplasmin's activity and genotypes in veinthrombus embolism.Methods: 95 normal controls and 102 VTE patients including 28 PTE patients and 74 DVTpatients diagnosed by ultrasonography and/or venougraphy were studied. Theα2-Antiplasmin activity (α2-AP: Ac) was measured among the patients and controlsusing one-stage clotting assay. Theα2-AP Arg6(CGG)/Trp(TGG) SNP genotype andthe allele frequency in the VTE group and the normal control group were determineby allele-specific polymerase chain reaction(AS-PCR).And the results were certifiedby sequencing.Results: 1.The plasma level ofα2-AP: Ac was elevated significantly in patients with VTE thanin controls (P<0.05). 2. The genotype distribution was in accordance withHardy-Weinberg equilibrium (p>0.05). The frequencies of CC genotype were59.8%and 64.2% in the VTE group and control group, CT 36.3%,28.4%, TT 3.9%,7.4%respectively. The frequencies of allele were C 77.9%,78.4%, T 22.1%,21.6%.Thegenotype and allele were not obvious statistics difference(p>0.05). 3. The plasmalevel ofα2-AP: Ac has no relationship with theα2-AP Arg6Trp genotypes (p>0.05).Conclusion: It suggests that theα2-AP: Ac level obvious markup in the VTE patients, it ispossibly one of VTE risk factors. The normal controls and VTE patients hadα2-APpolymorphism, but there was no significant relationship between the polymorphismofα2- AP and VTE. The plasma level ofα2-AP: Ac has no relationship with theα2-AP Arg6Trp genotypes.
