Objective: We aimed to explore the relationship between VTE and Fâ…§: Ag and vWF: Ag by detecting the Fâ…§: Ag plasma concentration as well as its vector of vWF: Ag plasma concentration and explore whether the Fâ…§gene 92714C>G polymorphism is an inherited VTE risk factors for Chineses patients, and learn more about the relationship between Fâ…§and VTE.Methods: 40 patients with venous thromboembolism from the First Affiliated Hospital of Dalian Medical University Respiratory June 2007 to October 2008 there is complete information as the experimental group, 40 cases divided into four groups:patients with DVT combined PTE group (n = 10) ; simple PTE group (n=10) ; chronic thromboembolic pulmonary hypertension (CTEPH) group (n=10); malignant tumor combined VTE group (n=10); and selected the healthy(n=10) and simple malignant tumor patients (n=10) in the same period whose sex and age matched with the VTE group and no history and a family history of thrombosis as normal control group and control group of malignant tumor combined VTE group respectively. After collecting blood samples, respectively, detected factorâ…§-related antigen (Fâ…§:Ag), human von Willebrand factor antigen (vWF:Ag) by EILSA; detected FVIII 92714C> G gene polymorphism by DNA sequencing.Results:1. The human factorâ…§-associated antigen (Fâ…§:Ag) results: PTE combined DVT Group, simply PTE group and malignant tumors combined PTE group are higher than those in healthy control group, the difference is of statistically significance(P<0.05); PTE combined DVT group is higher than simple PTE group, the difference is of statistically significance (P <0.05); CTEPH group is lower than other groups, the difference is of statistically significance(P<0.05); malignant tumor combined VTE group is higher than simple malignant tumor group, the difference is of statistically significance (P<0.05); the difference between simple malignant tumor group and CTEPH group is of no statistically significance (P>0.05); the difference between simply PTE group and malignant tumor combined VTE group is of no statistically significance (P>0.05).2. Human von Willebrand factor antigen (vWF:Ag) results: PTE combined DVT group and PTE group are higher than healthy controls, the difference is of statistically significance(P<0.05); PTE combined DVT group is higher than simple PTE group, the difference is of statistically significance (P <0.05); CTEPH group is lower than healthy controls group, PTE combined DVT group, simple PTE group alone, the difference is of statistically significance (P<0.05); malignant tumor combined VTE group is lower than the healthy control group and simple malignant tumor group, the difference is of statistically significance (P<0.05); the difference between simple malignant tumor group and the healthy control group is of no statistically significance (P>0.05); the difference between CTEPH group and malignant tumor combined VTE group is of no statistically significance (P>0.05).3. Sample of 19 cases of pulmonary embolism DNA sequencing of PCR products, only 1 case was found Fâ…§gene 92714C> G mutation, the other 18 cases weren't found Fâ…§gene 92714C> G mutation.Conclusions:1. The coagulation factorâ…§:Ag plasma concentration and venous thrombosis may be closely related, and showed that the level of Fâ…§may be be associated with the load of thrombosis.2. vWF and Fâ…§closely related, vWF and Fâ…§may participate in VTE formation together.3. The 92714C>G polymorphism of Fâ…§gene may involve in the pathogenesis of VTE, but the 92714C>G mutation may not be the Chinese Han patients genetic risk factors of VTE, suggesting that gene polymorphism may have ethnic differences.
