1. Clinical research1.1Objective:The study of children bronchial asthma in remission stage of syndrome distribution law, discuss the relationship between symptoms- syndrome, provide the basis for the diagnosis and treatment of children with asthma and TCM syndrome differentiation typing.1.2Method:According to the children asthma TCM syndrome investigation research and epidemiological studies, remission of the TCM syndrome questionnaire formulated in children with bronchial asthma. Investigation of children bronchial asthma in remission stage in 214 cases, mark card type frequency analysis, cluster analysis, principal component analysis and correlation analysis on the age, gender, family history of asthma, flu season, inducement, past medical history, symptoms, tongue and pulse.1.3Result:214 cases of children with asthma in remission 139 cases were male(65%), female 75 cases(35%), there is a family history of 173 cases(80.8%), the incidence in winter season onset in 120 cases(56%), in the spring of 56 cases(26.2%), predisposing factors in 126 cases of respiratory tract infection(58.9%), with a history of eczema of the 66 cases(30.8%).After the four diagnostic methods of TCM syndrome differentiation, 104 cases of spleen and lung qi(48.6%), 69 cases of lung and kidney yin deficiency(32.2%), 41 cases of spleen kidney yang deficiency(19.2%).1.4Conclusion:In 214 cases of children with asthma in remission stage: The sex ratio distribution is different, have a family history of asthma, winter good incidence, predisposing factors to respiratory tract infection more common, past many history of eczema. Syndromes associated with symptoms of Cough, catch a cold repeatedly, tongue and pulse fingerprint.2.Experimental study2.1Objective:Study on the function of the young asthma rats lung(Pulmonary function, PF), the parameters of the morphological changes of lung tissue and expression of PDGF, TGF- beta 1, c-fos in rat asthma model, To provide the clinical basis for the prevention and treatment of children asthma with Traditional Chinese Medicine.2.2Method:The rats of clean SD adolescent with 120 grades(95±5g weight), were randomly divided into normal group: 4 weeks(Z4)(Z8), 8 weeks and 12 weeks(Z12), model group: 4 weeks(M4)(M8), 8 weeks and 12 weeks(M12), Jian Pi Bu Fei Hua Tan Fang group :(J4) 4 weeks, 8 weeks(J8), 12(J12) and DXM group: 4 weeks(D4)(D8), 8 weeks and 12 weeks(D12), The total of 12 groups, 10 in each group. By intraperitoneal injection the mixture of 1ml/rat with gel of OVA(100 mg)+Al(OH)3(100 mg) of first day and eighth day to sensitization rats, trigger asthma with albumin solution atomization in 15 th day, give different drugs respectively starting from the 16 th day, according to the different time nodes, each group with regular feeding respectively according to 4 weeks, 8 weeks, 12 weeks and kill the rats a week later. With instruments and image analysis software, observed the pathological slices of each group rats, PEF, FEV0.2, FEV0.2/FVC and airway morphology parameters; Using reverse transcription polymerase chain reaction(RT-PCR) to detect the time Point of PDGF, TGF- beta1 and c-fos expression change; PDGF and adopts the linear correlation analysis of TGF-beta1 and c-fos, and the correlation of PDGF, TGF-beta1,c-fos and rat airway geometry parameters.2.3Result:The normal control group Compared with the asthma model group, lung function, airway morphology parameters, the expression of PDGF,TGF-beta1, c-fosm RNA by RT-PCR method, P<0.01;Jian Pi Bu Fei Hua Tan group and DXM group respectively compared with asthma model group,P<0.05; Jian Pi Bu Fei Hua Tan group respectively compared with DXM group by the indicators, there was no significant difference,P> 0.05;the protein expression of PDGF was significantly positively related to the protein expression of TGF-beta1,c-fos,P<0.01,and positive correlation between the protein expression of PDGF,TGF-beta1,c-fosand the airway morphology parameters of WAt/Pbm,WAi/Pbm and WAm/Pbm, P<0.01.2.4 Conclusion:2.4.1Jian Pi Bu Fei Hua Tan Fang can improve the PF and tissue morphological parameters of asthma rats.2.4.2The expression of PDGF, TGF-beta1 and c-fos in airway remodeling in rats were increased, after the use of Jian Pi Bu Fei Hua Tan Fang, the expressions of them were significantly reduced. It showed that PDGF, TGF-beta1 and c-fos play a role in airway remodeling of asthma, and Jian Pi Bu Fei Hua Tan Fang can cut through PDGF, TGF- beta 1,c-fos for the inhibition of airway remodeling.2.4.3The expression of PDGFm RNA was significantly positively related to the expression of TGF-beta1, c-fosm RNA and the expression of PDGF, TGF-beta1, c-fosm RNA were positive correlation with the airway morphology parameters of WAt/Pbm, WAi/Pbm and WAm/ Pbm. It Showed that the occurrence and development of asthma airway remodeling effects on the expression of PDGF, TGF-beta1 and c-fosm RNA.
