OBJECTIVE: To study the effect of experimental varicocele on intratesticular testosterone concentration (ITC), serum testosterone(T), follicle stimulating hormone(FSH), luteinizing hormone (LH) and androgen acceptor (AR), follicle stimulating hormone acceptor (FSHR), luteinizing hormone acceptor (LHR) in testis of adolescent rats.Methods: Establishing experimental left varicocele animal model by partial ligation of renal vein in male Wistar rats. 36 Wistar rats were randomly divided into 6 groups, EV continued 2 weeks, 4 weeks, 8 weeks respectively and the corresponding control group received sham surgery. 2 weeks, 4 weeks, 8 weeks after successful modeling, inferior vena cava blood and the testes of the EV models and controls were extirpated. Inferior vena cava blood was centrifuged and the serum was obtained. Take part of the testis and make testis tissue homogenate. The remaining testis was wax-embedded.The gonadal hormone concentration and ITC were studied using Radioimmuno- assay. The AR, FSHR and LHR were studied using immunohistochemistry. The apoptosis index was detected by TUNEL.Results: 2 weeks after surgery, there were no significant changes in ITC, scrum T, FSH, LH, but the expression of LHR increased remarkable(p﹤0.05). There were no significant changes in FSHR and AR. There is a remarkable increase of AI in leydig cells compared between the EV groups and the control groups. There were no significant changes in ultrastructure in EV groups and the control groups.4 weeks after surgery, there was a remarkable decline of ITC compared with the control group and 2-week group(p﹤0.05), but there were no significant changes in scrum hormone. There was a remarkable decline of the expression of AR than the 2-week EV group. There was a significant increase in testis germ cell apoptosis. There was significant damage in ultrastructure in EV groups8 weeks after surgery, there was a remarkable decline of ITC, scrum T, FSH and LH of the EV group compared with the control group, the 2-week EV group and the 4-week EV group. There was a remarkable decline of the expression of AR, FSHR and LHR compared with the control group, the 2-week EV group and the 4-week EV group. There was a significant increase in testis germ cell apoptosis of the EV group compared with the control group, the 2-week EV group and the 4-week EV group. The damage to the ultrastructure of seminiferous tubules was aggravated with the prolonging of varicocele.Conclusion: 1. Experimental varicocele decreases ITC, serum T and the expression of AR.2. Experimental varicocele affects the concentration of serum FSH, LH and the expression of their acceptor.3. Experimental varicocele increases the AI of germ cells and causes serious damage to the spermatogenesis.
