| Objective:To study the effect of Compound Salvia on canine tooth autotransplantation, and possible mechanisams of the Salvia on periodontal tissue of autotransplanted teeth.Material and Methods:Selecting five adult male hybrid dogs as experimental animals, weighing 10~12kg, which were randomly divided into two groups:the normal control group (2 dogs) and the experiment group (3 dogs). The dogs'teeth in the experiment group were autotransplanted and the dogs were fed Compound Salvia tablets,3 tablets per time,3 times a day. Dogs in the control group just underwent the teeth autotransplantation. On the first day, two teeth of each dog were pulled out and the teeth were autotransplanted into the contralateral corresponding mandibular alveolar fossa. The transplanted teeth and the adjacent teeth were ligated with the ligation wire∞. The same operation was carried out on the 10th,17th and 24th days and all dogs were killed on the 31st day. The transplantation time of the 5 dogs' teeth were thus divided into four periods:1 week after transplantation,2 weeks after transplantation,3 weeks after transplantation and 1 month after transplantation. After killing the dogs, making their maxillary and mandible fixed into neutral formalin. Then, with one tooth as a unit, the buccolingual tooth-periodontium was taken as the joint specimens, the size being about 1.5×1.0×0.5 cm. Put the specimens in the 10% EDTA decalcification solution which were placed in the microwave for the decalcification. The temperature was controlled between 55~65℃. The conventional dehydration, transparentizing, dipping into wax, embedding, slicing were performed after the decalcification. Later, the teeth-periodontal joint specimens of each group of transplanted teeth were stained with HE and immunohistochemical staining was done to them with the method of the two-step of the powervisionTM(PV) to detect the expression of TGF-β1 or PCNA. The average optical density was analyzed. The statistical software SPSS16.0 was used to analyze the result. As to the statistical method, the variance analysis of the data collected from the repeated measurements and comparing LSD pairwise and Pearson's correlation analysis were adopted. The testing standard a was 0.05.Result:(1) Compared with control group, the HE staining of the tissue sections of the experimental group showed more undifferentiated mesenchymal cells in the bone marrow cavity, more osteoblast at the bone trabecular edge, and more cementoblasts cells and fibroblast in the Periodontal ligament. They had large volume and large deeply-stained nuclear. There are more small blood vessels and obvious bone matrix generated. In the 1-4 weeks of the tooth transplantation, the remodeling of the periodontal tissue of the transplanted teeth is gradually active. While, for the control group, the undifferentiated mesenchymal cells in the bone marrow cavity and osteoblast at the bone trabecular edge are less, the volume is small,and bone matrix generated are less. (2) PCNA immunohistochemistry staining shows that there is positive expression in fibroblast,undifferentiated mesenchymal cells in the bone marrow cavity, undifferentiated mesenchymal cells aroud vessel, blood cells of vascular interior wall and osteoblast at the trabecular edge. There is occassionally positive expression in bone cells of the bone matrix. The positive expression of the experimental group is significantly higher than control group. There is a Statistical difference between experimental group and control group (P<0.001), and the positive expressions of the two groups both increase as time goes by in the 1-4 weeks. The average PCNA levels in the Periodontal tissue of the transplanted teeth in the 2nd week and in the 1st month are higher than that in the 1st week and in the 3rd week. The highest PCNA level is in the 1st month.(3) TGF-β1 immunohistochemistry staining shows that there is positive expression in undifferentiated mesenchymal cells in the bone marrow cavity, undifferentiated mesenchymal cells around vessel, blood cells of vascular interior wall. There in no expression in mature bone cells.There is positive expression in fibroblast in the periodontal ligament and extracellular matrix. The positive expression of the experimental group is significantly higher than the control group. There is a Statistical difference between experimental group and control group (P<0.001) and the positive expressions of the two groups both increase as time goes by in the 1-4 weeks. The expression is the highest in the 3rd week in experimental group. Conclusion:PCNA and TGFβ1 participates in and contributes a lot in the remodeling of the transplanted tooth periodontal tissue. The positive expression of PCNA and TGF-β1 in the transplantation of tooth periodontal tissue have not found anything in correlation. Maybe, by increasing the expression of PCNA and TGF-β1 in periodontal tissues, the Salvia promotes the proliferation and differentiation of periodontal ligament cells, promotes the periodontal tissue remodeling. |
