Objective: Glioma is the most common brain tumor with the incidence of 40.49%. Malignant glioma is characterized with infliltrating growth without boundary between the normal brain tissue.Malignant glioma betides several brain lobes and is difficult to be ablated totally. Each part of glioma has different pathological characterization and different malignant level. Glioma cells are insensitive for radiation and chemotherapy drugs are hard to permeate blood-brain barrier.The results of radiotherapy and chemotherapy are not satisfactionary after operation. The recurrence rate after surgerical operation is high and the median survival is 14.6 months. Deep research has discovered that turmor stem cell is correlated with the glioma recurrence, and the brains turmor stell cell hypothesis is proposed.Brain tumor stell cell (BTSC)theory suggested that there are a small part of cells in the tumor tissue with stem cell characteriztion has the ability of self-renewal and multilineage differentiation,which is the source of the formation of tumor cells with different degree of differentiation and the tumor promotion. Currently,CD133 and Nestin are specific markers of BTSC. PCNA , the tumor indicator, has been used in the proliferational evaluation.The study is aimed to investigate the BTSC proliferation through CD133?PCNA and Nestin?PCNA coexpression in different grade gliomas,and the influence of the BTSC to the proliferation of tumor has been explored.Method: 95 cases of Human glioma were detected by Sp immunohis ochemical method. 62 cases with CD133 expressed were adopted into the test in order to investigate the CD133/PCNA coexpression.On the other hand, 70 cases with Nestin expressed were adopted in order to investigate the CD133/PCNA coexpression. CD133/PCNA and Nestin/PCNA coexpression in different pathological grade gliomas was investigated with immunohistochemistry double staining method and their relationship was also analyzed. Statistics:Spss13.0 statistics software was used to data analysis, and numeration data was measured withχ~2 analysis, P<0.05.Results: According to the separate criteria of nervous system neoplasms,the results as follows:21 cases in the low grade(Ⅰ,Ⅱ). 41 cases in the high grade(Ⅲ,Ⅳ).The Nestin positive example:23 cases in the low grade, 47 cases in the high group, In the CD133/PCNA positive group, 21 cases was in the low group,9 cases was C/P coexpression,41 cases was in the high group,30cases was C/P coexpression, and the positive rate were 42.86% and 73.17% respectively.χ~2=5.469, P=0.019.The difference was statistically significance. In the Nestin/PCNA positive group, 23 cases was in the low group,11 cases was N/P coexpression, 47 cases was in the high group,38cases was N/P coexpression, and the positive rate were 47.62 % and 80.85 % respectively.χ2=8.020, P=0.005.The difference was statistically significance. All the results indicated that significant differences existed in different pathological grade of glioma group.Conclusion: The results showed that the increased expression of CD133/PCNA and Nestin/PCNA in glioma along with the malignancy, and there was significant differcence between the control group. In tissue of glioma cells with CD133 positive expression than the cells with Nestin expression has more specificity relatively. Immunohistochemistry double staining of CD133/PCNA and Nestin/PCNA is in favour of judging glioma biological behaviour more accurately and assessmenting patient prognosis,which could provide experimental bases for targeted therapy directed specially and exactly to stem cells of tumor .
