| Objective As genetic markers, short tandem repeats (STR) is widely used in personal identification and parentage testing. Up to now, more and more STR loci have been developed which are more powerful in forensic practise than some loci used now. It is needed and feasible to develop new amplification kit. AmpFlSTR? SinofilerTM PCR Amplification Kit is specially developed for Chinese forensic laboratories, in which D6S1043 and D12S391 are substituted for TH01 and TPOX in identifiler kit. Other loci included in the kit are D8S1179, D21S11, D7S820, CSF1PO, D3S1358, D13S317, D16S539, D2S1338, D19S433, vWA, D18S51, D5S818 and FGA. However, there are little population-genetic indices about this kit as a whole. In order to evaluate this kit and to get basic population-genetic indices for its use in forensic practice in Chinese Han population, the investigation was carried out.DNA isolated from 231 unrelated Han individuals from Henan were typed using the Sinofiler Amplification Kit according to the manufacturer manual. Allele frequencies and other forensic indices were calculated using powerstat software. Exact test for Hardy-weinberg Equilibrium (HWE), linkage equilibrium and comparison between different populations were conducted Using Arlequin woftware.1. Population variation and forensic indices:The most discriminating locus was D6S1043 while the least was D3S1358. The combined match probability was 9.81×10-19 and the combined power of exclusion was 0.99999974. Statistical analysis of the generated data indicated no departure from expectation of Hardy-Weinberg Equilibrium (HWE) in all loci but D6S1043. The observed and expected heterozygosity, power of discrimination, polymorphic information content, the other population-genetic indices were calculated.2. Exact test for linkage equilibrium were performed in all pairs of loci using Arlequin software. The p value of 9 out of 15 locus pairs were more than 0.05, while that of 6 pairs (D12S391 and D19S433,D12S391 and D5S818,D13S317 and D2S1338,D16S539 and D21S11,D19S433 and D5S818,D3S1358 and D8S1179) were less than 0.05, but the p value were not signifcance after Bonferroni's correction. The p values for linkage equilibrium of D18S51and D2S1338 were signifcance even after Bonferroni's correction.3. There were more genetic difference between Henan Han and Han from southern part of China than between Henan Han and Han from northern part of China.4.Thirteen rare alleles had been found from D16S539(1), D21S11(2), D7S820(1), D6S 1043(4) and FGA(5) most of which were microvariant, with a allele frequence of 0.0022 to 0.0087.1. Sinofiler PCR amplication kit can be used in forensic practice in Han population. It is more powerful than identifiler kit when used in personal identification and parentage testing.2. The population indices in Han population, which are essential for its use, were collected. As no difference of allele distribution between Han population from different aera of northern China, the data can be used not only in Henan Han, but also in other northern aera of China. |
