Hydrogen Selenide, a kind of gas with higher toxicity than sulfuretted hydrogen, packs quite a punch on upper respiratory tract mucosa and conjunctival. The persons who touch the gas within several minutes to three hours will appear toxic symptoms in succession. And the serious patients may further develop into chemical pneumonia or pulmonary edema.The toxicity of each chromium compound is very different. It packs a punch on mucous membrane and may bring dermatitis and damages on liver and kidney. And the hexaalent chromium compounds have the cancer-sensitized effect. The serum chromium can reflect the body healthy level and situation of touching soluble chromium, which is an important biological supervising index for the persons who are lack of chromium or the professional and unprofessional contactees.From the above reasons, it can be seen that sampling of hydrogen selenide in the air of working sites and chromium trace determination in human's serum are very important. The atomic fluorescence spectrometry for hydrid has such advantages as rapidness, sensitivity, correctness, good choice, less disturbance and convenient operation, etc.And for the chromium that is hard to form hydrid it is better to use furnace atomic absorption spectrometry. This thesis is to research and set up sampling and inspecting methods of hydrogen selenide in the air of working sites as well as the determination of chromium trace in human's serum on the basis of the achievements made by predecessors.The paper mainly focuses on the following research:1,Study on Atomic Fluorescence Spectrometry to Determine Hydrogen Selenide in the Air of WorkshopsIt is to set a sampling and inspecting method of hydrogen selenide in the air of working sites, which applies lOmL 0.1mol·L-1 NaOH as the absorption liquid and 0.1L·min-1 gas production flow velocity. This thesis analyzes such aspects as the order of acid-base titration to create Hydrogen Selenide, proportion and consistency of Hydrogen Selenide, producing conditions of Hydrogen Selenide, digesting conditions of absorption liquid, gas production flow velocity and choice of absorb liquid, etc., which optimizes the conditions of the instrument, including consistency of HCl carrier and consistency of NaBH4 & NaOH liquor, etc.Under such optimized experimental conditions, the standard curve scope of hydrogen selenide was 4.0-200μg·L-1,liner correlation was 0.9999,detecting limit was 4.0μg·L-1 and the addition standard recovery was 94.2%~105.6%,which can satisfy the requirements of determination of hydrogen selenide in the air of working sites.2,Study on Furnace Atomic Absorption Spectrometry to Determine Chromium in SerumIt is to set up a new method to inspect the chromium trace in human's serum, which mainly researches the improvement of the analyzing methods of chromium trace in serum by using of Mg(NO3)2 as the Matrix Modifier and optimizes the conditions of choice of acid and acidity, adding amount of Matrix Modifier and choice of atomization and ashing temperature, etc.Under such conditions,the detecting limit was 0.07μg·L-1,relative standard deviation was 2.23%,actual recovery was 92%~105%.Because the samples do not need complex pre-chemical treatment and can be detected, which is simple, convenient, fast, exact and accurate, this method is worth being popularized widely.
