Objective To observe the cure effect of Jianxizhuanggu medication on rats knee osteoarthritis and explore the treatment mechanism. Provide experimental basis for the development of new drugs of TCM.Methods 60 rats were divided into normal group (A) 10 surgical group 50 randomly. rats knee osteoarthritis model was induced with the improved method of Hulth. Surgical group was divided into control group B, Zhuangguguanjiewan control group C, High dose Jianxizhuanggu group D, moderate dose Jianxizhuanggu group E and low dose Jianxizhuanggu group F.7 days after model induced, give 5ml drugs to the rats of Jianxizhuanggu group. It equals to 8g,4g, and 2g pills. Give 5ml normal saline to the rats of Zhuangguguanjiewan control group. It equals to 2g pills. Give 5ml to the rats of normal and model group. Rats were killed 1 and 2 months later each a half. Take cartilage of lateral femoral condyle to measure apoptosis of cartilage cells and regulation of gene expression P53 and Bcl-2.Results Experiment group demonstrates the ability to convert the apoptosis trend of the cartilage cells compared to other groups. It is superior to other groups in down regulation the hyperexpression of gene P53 and turning up the suppressed Bcl-2 expression. There is significant difference (P<0.05).Conclusions Cartilage apoptosis is one of the important pathogenesis of osteoarthritis. Janxizhuanggu medication can convert the apoptosis trend of the cartilage cells. This may relate to the influence of gene expression P53 and Bcl-2 which protect cartilage cells to some extent and prevent development of osteoarthritis.
