The prescription of double coptis preparations include the skullcap, honeysuckle and forsythia, it possess expelling wind and diaphoresis relieving superficies,heat- clearing and detoxicating, for the treatment of influenza caused by exogenous wind-heat. Forsythia volume in double coptis preparation is the sum of the remaining two. Forsythiaside and Phillyrin in Forsythia as characteristic components of these two components of which control more meaningful. Forsythiaside is so far in the strongest component of antibacterial activity from Forsythia species.Therefore it is more suitable to increase the evaluation of the compound when evaluating the efficacy of antimicrobial agents as the main quality. But the quality of evaluation of such agents mostly phillyrin as the index now, which is clearly inappropriate.So it is necessary that establishing the assaying method accurately, simplely and reproducibly.This topic conducted a qualitative an quantitative research about the active ingredient of forsythia in Double coptis preparations.In this study, the forsythia medicinal active ingredients Forsythiaside and Phillyrin were conducted quanlitative identification by TLC. The results show that identification method is simple, clear spots, good reproducibility of the preparation,and can effectively identify forsythia medicinal herbs.In this study, the forsythiaside and phillyrin of extraction method was improved by using orthogonal test method involves investigating the factors in extraction process in Double coptis preparation, and we established the optimal extraction program,that is the sample is dissolved with 25mL 50% methanol, ultrasound 20 minutes.In this study, the assaying of forsythiaside and phillyrin of forsythin in Double coptis preparations was determined by high performance liquid chromatography. The results show that Forsythiaside has a good linear relationship in 3.22-128.8mg/L range the correlation coefficient was 0.9999 (n=6);Forsythin has a good line in 0.99-39.6mg/L range and the correlation coefficient was 0.9999 (n=6). The method is simple, high sensitivity,good reproducibility,and it provided a quantitative basis for the control of the quality of forsythia medicinal in Double coptis preparations.This study is based on the results of determination about forsythiaside and phillyrin,and developed concentration limits on forsythiaside and phillyrin in a series of Double coptis preparations. The measured results showed that in the preparation of different manufacturers, Forsythiaside and Phillyrin were significantly different, especially the differences of forsythiaside was larger.The common oral liquid dosage forms in a series of Double coptis preparation was used in for the study, and we collected eight manufacturers of more than ten batches of samples to conduct fingerprint. The similarity of 10 batches of Liquid samples was above 0.90, and it indicated the double coptis different manufacturers between different batches have small differences and stable quality.The results of this research show that: in the preparation of different manufacturers, Forsythiaside and Phillyrin were significantly different, especially the differences forsythiaside relatively large,.To some extent, it explains the quality control standards on double coptis in the current pharmacopoeia only for control of baicalin is not enough, and the forsythiasi- de content limit of standard in forsythia medicinal herbs should be established,it will play a crucial role on improving clinical efficacy and quality of this preparation in the future.
