Scorzonera divaricata was investigated in this article. The key content were that extraction technology and antioxidant activity of total flavonoids in S.divaricata, and the seasonal change rule of the content of total flavonoids and phenol. Meanwhile, isolation and purification of ethanol extraction of S. divaricata were also studied and a monomeric chemical substance was identified. And here were as follows:1. A plant chemical composition systemic study method was used to preliminarily identify the kinds of chemical compounds in S. divaricata.2. Extracting technology of total flavonoids in S. divaricata was optimized by Uniform Design. The results showed that the hot reflux extraction and ultrasonic-microwave assisted extraction of total flavonoids in S. divaricata were better compared with ultrasonic-assisted extraction, soaking and shaking extraction and microwave-assisted extraction. Their optimum extraction conditions were as follows: extracting time was 45 min, the ratio of material to liquor was 34:1, concentration of ethanol was 15% ( the hot reflux extraction ); extracting time was 41 min, the ratio of material to liquor was 36:1, concentration of ethanol was 30%, extracting times was 3 ( ultrasonic-microwave assisted extraction ).3. The results of antioxidation in vitro of total flavonoids from S.divaricata showed that the total flavonoids had a strong antixidation. The ability of antioxidation become stronger along with the increase of concentration of total flavanoids, having a certain concentration-response correlation. During the measuration of antioxidative activity against DPPH, the dynamic curve, Y= (Y0-NS)×exp(-K×X)+ NS, can well show the dose-effect relationship.4. The research results of anti-lipid peroxidation of total flavonoids from S. divaricata showed that extractions had a certain inhibitory action against Lard, but are not suitable to be added as antioxidant of rapeseed oil, sunflower oil and cottonseed oil.5. Every July and August is the best time to pick S. divaricata in Xingjiang to study the total phenols and flavonoids from it..6. Isolating and purifying repeatedly the ethanol extraction of S. divaricata, gained a monomer finally, and it present white crystal in chloroform-methanol. The name of this crystal is betuli through structural identification.
