This thesis mainly can be divided into two parts,the first part is the isolation and Characterization of Chemical Constituents from the fresh roots of Raphanus sativus L,the second part for the study of the extraction process of peony petals.ObjectiveChinese white radish(Raphanus sativus L.)is a traditional vegetables and herb of the Brassica genus for more than 1000 years in china.It has shown anticancer?antioxidant and treatment of gastrointestinal diseases.in modern pharmacology.However,because it has few report in the chemical composition of radish,in particular its essential oils.So It is not clear in the efficacy of the chenical composition.Wo hople to explicit material basis of efficay in order to provide the basis for research and development.paeonia ostii flower is Ranunculaceea peony plants,also known as "rich flower ",is our specialty important ornamental plants and medicinal plants.The root of paeonia is the source of common DanPi medicine.Paeonia.paeonia ostii flower in addition to ornamental value,other economic value has untapped potential.Recently Paeonia ostii T.Hong et J.X.Zhang has been approved by the State as new food material(the food of new resource)in the December 2013.We study the extraction of Paeonia.paeonia ostii flower in order to used as new raw ordinary food,health food raw materials,cosmetics raw materials,further study medicine of dysmenorrheal treatment.Methods and ResultsThe fresh roots of Raphanus sativus L.(80kg)were extracted with water four times under reflux.After removing the solvent under reduced pressure at 65?.The concrete was suspended in distilled water and extracted with three times Petroleumether,ethylacetate,and n-butanol,consecutively.The Petroleum ether extract was chromatographed over a silica gel column and eluted with a sytem of Petroleum ether and ethyl acetate to furnish three compounds:compound I(30mg),compound ?(20mg),.The ethylacetate extract was chromatographed over a silica gel column and eluted with a sytem of CH2Cl2:CH3OH to furnish compound ?(20 mg).The n-butanol extract was chromatographed over a silica gel column and eluted with a sytem of CH2Cl2:CH3OH to furnish three compounds:compund IV(8g),compound V(20 mg),and compound VI(20 mg).and In the present study,6 compounds were isolation and characterized from water extract of the.Raphanussativus L,these compounds were elucidated asp-sitosterol(?),,ursolic acid(?),berberine(?),Daucosterol(?),n-butyl-?-D-fructopyranoside(V)among these compounds,were all isolated from Raphanussativus L.for the first time and compounds ?,?,?,? were first reported from the family of Cruciferae.Volatile oil extracted by water steam distillation and analyzed by GC/MS,and theirrelative contents(in%)were found by the method of normalization.There were 42 chemical compositions are identified by GC/MS method.The main compounds were 1,14,17-Eicosatrienoicacid,methyl ester,Dimethyl trisulfide,n-Hexadecanoic acid,9,12,15-Octadecatrienoic acid,methyl ester,(Z,Z,Z)-,Tetrasulfide,dimethylPeony alcohol extraction process of main to extract the flavonoids and anthocyanin in indexprocess.Flavonoids eventually chose AICl3-CH3OOK-complex legal measure metal ions and health care products determination method of total flavonoids,anthocyanins determination method to select the reference substance and PH color difference method.Technology is investigated extraction method,extraction solvent,solvent volume fraction,the liquid ratio,ultrasonic frequency four factors,in combination with 34 orthogonal experiment to optimize the extraction process.Is A3B1C2D2 with the extraction process of total flavonoids as index,the extraction process of anthocyanins as indicators for A2B1C3D2,and extraction technology were determined by four kinds of design and color of peony petals of flavonoid and anthocyanin content was followed by Dan peony fung reddish flavonoids content of 1.33%,anthocyanin content was 0.154%;Dan peony fung red outside the flavonoids content of 2.12%,anthocyanin content was 0.174%;Dan peony fung purple color outside the flavonoids content of 2.02%,anthocyanin content was 0.778%;Yellow Dan peony fung flavonoids content 1.68%,anthocyanin content was 0.083%.This chapter mainly studies the extraction and purification technology of essential oil.Essential oil extraction technology of essential oil fragrance for sensory index and extract rate for precise index to study the extraction process of essential oils,the selected extraction solvent,grinding time,liquid ratio,timeout four factors,the single factor investigation,and ultimately chose the water as the solvent,liquid ratio,grinding time,ultrasonic time,three factors made orthogonal experiment to optimize the process,finally determine the essential oil extraction technology for taking water as solvent,the extraction process of A1B2C3.Essential oil purification process was investigated biology methods,physics method,organic solvent method purifies the comparison,finally chose the organic solvent method,comparing the several kinds of organic solvent method purifies the essence of life,finally chose the steam distillation extraction solvent,liquid ratio,extraction time,three factors,and through the orthogonal experiment to optimize the process eventually determine the essence of pure method for A1B2C2,is extracted by the extraction and purification technology of essential oil with flowers and flower extract the volatile oil,and its volatile components were determined by GC-MS analysis.Flowers in the 32 g,essential oil content is 0.45%,the peony 41 g.The essential oil content is 0.256%,peony essence oil is pale white,more has full-bodied fragrance,density<1 g/ml.The essential oil by GC-MS determination of volatile oil components,respectively,from the peony flower essential oil compounds isolated from 27,were detected with 22 kinds of compounds;Isolated from peony extract of 24 kinds of composition,a total of 19 species composition.Can be seen in the peony flowers to contain terpenes,esters,ketones,acids and rich hydrocarbon compounds.Peony extract contains terpene,esters and abundant hydrocarbon compounds.Both identified compounds are basically the same,in the flowers 4-phenyl-4,4 a,5,6,7,8-six hydrogen-2(3 h)-naphthalene,and cuckoo ketone,and there are no identified in peony extract,identified from the peony extract ethyl oleate.The peony extract temperature,illumination,oxidant and reducing agent was studied,the influence of metal ions on its stability,the results showed that the peony extract relatively stable at 4?,25? is the most unstable,40? is relatively stable,low light influence on the stability of the peony pigment.Hydrogen peroxide oxidant and reducing agent sodium sulfite to how great the influence of the peony pigment,and then the air.Metal ions,magnesium 2+Na+,K+in two days of peony extract stability,Fe3+,magnesium 2+,Cu2+ on peony extract is very unstable.More than 3 days after the absorbance increases gradually,is likely to be metal ions and the inside of the water extract ingredients generated the complexation ion,lead to absorbance increase.According to the 2010 edition of pharmacopoeia method for determining the peony and peony extract of ash content,peony ash content is 5.76%,acid insoluble ash content is 0.76%,the peony extract ash content is 2.27%,acid insoluble ash content is 0.24%.ConclssionIn the present study,6 compounds were isolation and characterized fromwater extract of the.Raphanussativus L,these compounds were elucidatedas?-sitosterol(?),ursolic acid(?),berberine(?),Daucosterol(?),n-butyl-?-D-fructopyranoside(?)among these compounds,were all isolatedfrom Raphanu ssativus L.for the first time and compounds ?,?,?,? were first reported from the family of Cruciferae.And we forest report the components of Volatile Oil In the fresh roots of Raphanus sativus L.We has established the extraction and measured methods of Paeonia ostii flowerin the flavonoids and anthocyanins.And we established a way for the extraction and purification the essential oil of Paeonia ostii flower.We established a method for the determination of Cyanidin-3-O-glucoside chloride by HPLC.And analysised the components of Volatile Oil In Paeonia ostii Flower and its exection by GC-MS.
