Objective:to investigate the influence of Berberine onβ3-cell on Fasting blood glucose (FBG),fasting blood glucose (FINS) and pancreatic structure in streptozotocin-induced type 2 diabetic rats.Methods:DM models were induced by streptozotocin (STZ)injection.DM models were divided randomly into 4 groups according therapeutic doses: high-dosegroup(180mg/kg),middle-dose group (80 mg/kg),low-dose group (40 mg/kg),diabetic control group,and set normal group, rats of normal group and diabetic control(DM)group received no drug,3 treatment groups were administered with berberine per day for 8 weeks, respectively. Rats were fasted for 10 hours,under anesthesia with chloral hydrate, blood samples were collected via the Abdominal aortic for detected FBG, FINS and calculate Insulin sensitivity index (ISI); groups were killed and pancreatic structure was observed with hematoxylin& eosin(H&E) staining,insulin(INS)and glucose transporter-4(GLUT-4) protein expressions were detected by immunohistochemistry.Results:(1) compared with normal group,FBG and FINS of model group was significantly higher(p<0.01); compared with DM group,FBG and FINS of low-dose group, middle-dose group and high-dose group all decreased (p<0.01); compared with middle-dose group and low-dose group,FBG and FINS of high-dose group decreased significantly (p<0.01),but had no different between middle-dose group with low-dose group (p>0.05).(2) compared with normal group,ISI of model group was significantly lower (p<0.01); compared with DM group, ISI of low-dose group, middle-dose group and high-dose group all increased significantly (p<0.01); compared with low-dose group and middle-dose group,ISI of high-dose group increased significantly (p<0.01),but had no different between low-dose group with middle-dose group (p>0.05).(3) compared with normal group,INS of model group was significantly lower (p<0.05); compared with DM group, INS of high-dose group increased significantly(P<0.001),but had no different with low-dose group and middle-dose group(p>0.05); compared with low-dose group and middle-dose group,INS of high-dose group increased significantly (p<0.05), but had no different between low-dose group with middle-dose group (p>0.05).(4) compared with normal group,GLUT-4 of model group was significantly lower(P<0.05); compared with DM group, GLUT-4 of low-dose group and middle-dose group increased significantly(P<0.05), but had no different between high-dose group(P>0.05); compared with low-dose group and middle-dose group,GLUT-4 of high-dose group increased significantly (p<0.05),but had no different between low-dose group with middle-dose group (p>0.05).
