Objective: To study the protein expression of Gold Capsule on phosphatidylinositol-3-kinasem,glucose transporter 4 in liver and skeletal muscle tissue of Diabetes mellitus and insulin resistance rats, and its change on blood glucose, body weight, insulin sensitivity index.Demonstrates the mechanism of Gold Capsule to improve insulin resistance and decrease blood glucose, in order to provide theoretical basis for Jieduhuazhuo Method of Traditional Chinese Medicine on Diabetic mellitus and insulin resistance from the cell level.Methods:Select 65 SPF level of male wistar rats( 4 months, 200-250g), to fed the basal diet and adaptive feeding one week, they were all in the standard ranges of body weight and blood glucose, 60 rats were brought into finally. Randomly selected 12 rats as the control, and the remaining 48 as a model group, after high-fat diet for 4 weeks and not provide diet more than 12 hours, 1% streptozotocin 50 mg.kg-1 were given into the rats by intraperitoneal injection, to build the rat model of Diabetes mellitus. After 72 hours, blood was collected from caudal vessel and OGTT. The diabetic rats were randomly divided into gold capsule group(gold capsule suspension, 2.025g/kg), rosiglitazone(rosiglitazone suspension, 0.5mg/kg), model group(normal saline, 1ml/100g), continued high-fat diet feeding, were treated by infusing the stomach with the medicine and blank group(normal saline, 1ml/100g), observe the general state of the rats, body weight and food intake, 10 weeks after the intervention,intraperitoneal injection 10% chloral hydrate to anesthetized rats, test FBG, FINS, ISI. and apply the method of immunochemical to detect the pathological changes, PI-3K and GLUT4 protein expression in rat liver and skeletal muscle tissue. All data using SPSS 17.0 software for statistical analysis.Results:1. After drug intervention 10 weeks, gold capsule group compared with the model group, body weight was significantly lower, the difference was statistically significant(P<0.05).2. After drug intervention 10 weeks, detect and compared with the FBG, FINS, ISI of rats, The results show that compared with the model group, gold capsule group and rosiglitazone group can reduce the Level of FBG and FINS of Diabetes mellitus and insulin resistance rats, increase the level of ISI. the difference was statistically significant(P<0.05); Compared with gold capsule group and rosiglitazone group, showing that the efficacy of each index in the rosiglitazone group was better than that in the gold capsule group, and the difference was statistically significant(P<0.05).3.the results of immunohistochemistry:the blank group has the strongest and the most obvious expression compared with other groups of PI-3K, GLUT4 protein expression in liver and skeletal muscle tissue(P<0.05). Model group showed low expression, the expression intensity lower than rosiglitazone group and gold capsule group(P<0.05), compared to rosiglitazone group and gold capsule group, the rosiglitazone group expression intensity was more stronger(P<0.05).Conclusion:1.The gold capsule developed by Jieduhuazhuo Method of Traditional Chinese Medicine, it can improve ISI, reduce body weight and blood glucose of Diabetes mellitus rats model.2. Gold capsule has the effect of improve insulin sensitivity, the mechanism is related with the activation of PI-3K and GLUT4 protein expression in skeletal muscle tissue insulin signal pathway.
