Objective Polymorphism of vascular endothelial growth factor receptor 2 (VEGFR2) influences the function of VEGF, which implicates a putative angiogenic basis in various types of diseases, but its role in the development of nasopharyngeal carcinoma (NPC) is still not fully understood. We sought to investigate whether the polymorphism of VEGFR2-604T/C was associated with susceptibility and invasion of NPC in Chinese population. Search and study the susceptibility of NPC gene and invasion metastasis-related molecular. In order to study the genetic mechanism of NPC and reveals the development of the internal factors and provide a theoretical basis for individualized treatment.Method We investigated the associations of VEGFR2-604T/C polymorphism with the susceptibility and clinical tumor characteristics of 172 NPC cases and 184 controls. The polymorphism were genotyped using real time-PCR technique in blood genomic DNA of all the subjects. Logistic regression analysis was employed to assess the associations between genotypes and the risk of NPC, adjusting for multiple confounding factors.Results The polymorphism of VEGFR2-604T/C was in accordance with Hardy-Weinberg balance law. It occurred three genotypes: TT,TC,CC. The three genotype of NPC patients and healthy control group had no significant difference (p>0.05). Stratified analysis by sex was not statistically significant. NPC patients according to clinical and pathological data were stratified for: The group of N0-2 about the VEGFR2-604T/Cand VEGFR2-604T allele frequency was significantly higher than N3 (p=0.016 and 0.013),â… ~â…£a phase group, p values were equal to 0.037; VEGFR2-604T/C was not significantly correlated with pathologic type, distant metastasis, gender, the age, nationality and family history of cancer.Conclusions In this study, VEGFR2-604T/C polymorphism and genetic susceptibility of nasopharyngeal carcinoma in Chinese population no significant correlation, and tumor biology behavior, including T stage, histological type,distant metastasis had no significant connection. In this study, we found that VEGFR2-604T/C was significant correlation between with N stage and clinical stage which carrying C allele may be a protection factors for invasive of NPC. In the clinical treatment will contribute to individualized treatment of nasopharyngeal carcinoma and may play an important role in the progression of the disease. The precise mechanisms of the polymorphism on the differing status in NPC should be elucidated in further studies.
