Objective:Osteosarcoma is a common malignant tumor, is one of the malignant tumor which can hazard to human health. The age was mainly children and adolescents, and the five-year survival rate was low, and prognosis was poor. The reason was that tumor may be have hematogenous microscopic metastasis, especially jumped metastasis when got the diagnosis of osteosarcoma, and the patients often occur the chemotherapy after operation.So to predict metastasis of osteosarcoma and evaluation of drug resistance become a research hotspot. Metastasis associated gene 1 (metastasis associated 1, MTA1) was upregulated in many malignant tumors, and was related with development and progression of tumor, especially the invasion and metastasis. The research show that the expression of multidrug resistance protein P-glycoprotein (P-gp) and DNA topoisomeraseâ…¡(Topoâ…¡) was related with occurrence of multidrug resistance mechanisms. In this study, we investigate the relationship between invasion and metastasis and drug resistance through the research of expression of MTA1,P-gp, Topoâ…¡protein in osteosarcoma after chemotherapy and hope to provide a basis of molecular biology for postoperative treatment of and prognostic evaluation of osteosarcoma.Subjects and methods:30 caseds of samples were collected from 2006-2007 about osteosarcoma after chemotherapy.In these samples to meet the following conditions: Preoperative chemotherapy for two courses, osteosarcoma was diagnosed by pathologists for after postoperative and followed. EnVision method of immunohistochemistry was used to detect expression of MTA1 and P-gp, Topoâ…¡protein in 30 osteosarcoma.Results:The positive rate of MTA1 and P-gp, Topoâ…¡protein expression was 60.0%,63.3% and 26.7% respectively. The positive expression of MTA1 was related to tumor metastases, exist or not of patients (P<0.05), but not with age, sex, malignant degree and position of tumor and Enneking staging (P>0.05). The positive expression of P-gp was related to malignant degree and metastases of tumor, exist or not of patients (P<0.05), but not with age, sex, position of tumor and Enneking staging (P>0.05). The positive expression of Topoâ…¡was related toexist or not of patients (P<0.05), but not with age, sex, position of tumor, malignant degree and metastases of tumor and Enneking staging(P>0.05).The expression of MTA1 and P-gp, Topoâ…¡was significant correlation in osteosarcoma after chemotherapy (P<0.05)Conclusions:The expression of MTA1 and P-gp, Topoâ…¡are closely related with the prognosis of osteosarcoma, and the three proteinum are combinedly detected that will evaluate effectivly the prognosis of osteosarcoma. MTA1 may be considered as molecular markers for prognosis of osteosarcoma and a target to molecular targeted therapy of tumor in the future.
