Objective: To observe the acute and chronic blood loss on coagulation and fibrinolysis system in the SD rat plasminogen activator inhibitor -1 (PAI-1) effects.Methods: SD rats were randomly divided into three groups (10 animals in each group): acute blood loss group(group A) and chronic blood loss group(group B), control group(group C). Group B rats were picked the blood every day one time. Before picking the blood, to be first clean the rat tail, starts from the tail terminal each time to cut approximately 0.5mm and each time picks blood 1.5ml. At the first day, group A rats were cut tail and picked blood 4.0ml (picked the blood the same way as before). The groups A and B were made into anemia model and continue picked blood 1.5 ml everyday. Group C rats were only cut tail and picked the oozing blood of tail. The groups A, B and C were observed eight days and not took any intervention measures during the period of picked blood .The picked blood was placed in a 5ml purple vacuum blood collection tube with EDTA. HB was detected and recorded.The blood samples were placed in 4℃, 4000 r / min, TGL-16G high-speed desktop refrigerated centrifuge centrifuging 10min.After that, plasma was collected EP tube, -20℃preservation. Take the blood plasma 100μl to examine PAI-1 with the ELISA test and observe the reducing of HB and content changing of the PAI-1 of each group.Results: The PAI-1 level increased in plasma when blood lost and HB decreased, and there is significant difference between control and blood loss groups. In group B, the content of PAI-1 in plasma increased markedly at first, and lastly maintained a high level with the increase in blood loss and decrease in HB. In group A, the level of PAI-1 reached a peak and fell rapidly, then increased slowly, and the range of increase was lower than that of group B.In group A, the PAI-1 level increased sharply at the first day of blood loss and reached the peak.It dropped fast from the second day , fell into the normal level in the 3rd day and latter raise to the higher level in a steady way, but not much higher than former high-value. In group B,the PAI-1 level gradually raised to reach a high value at the 3rd day and maintained one or two days at the high value .It declined slightly latter, but still maintained at the high level to the seventh day. However, the raising lever of group B was far higher than that of group A.It stated that the cruor-fibrinolysis system occured serious disorder at the 3rd day of blood loss. Such as: with the PAI-1 content advances, blood clotting strengthen.It may lead to DVT.Conclusions: The level of PAI-1 increased in plasma with blood lost and HB decreased.Two or three days after chronic lost blood, it was the critical period that body to adjust coagulation and fibrinolysis system. It also was the active period of thrombosis and bleeding tendency. That suggesting that more attention should payed to adjust coagulation and fibrinolysis system after 2-3 days of surgical trauma. It is the best time to prevent and treatment of deep venous thrombosis and occult blood lost. In the third day of acute blood loss, body almost reached normal lever .But after that, cruor-fibrinolysis system suddenly disordered. So 2-3 days after acute blood loss, it was the best time to prevent and treat the deep venous thrombosis and occult blood loss.
