Purpose:To investigate the expression and location of extracellular matrixmetalloproteinase inducer (CD147/EMMPRIN )and matrixmetalloproteinase-9 (MMP-9)in human salivary gland tumors,and the correlation between CD147 and MMP-9,andto evaluate the effect of CD147 and MMP-9 in soft and neural invasion.Methods: We detected the expression and location of CD147 and MMP-9 byPV-9000 polymer detection system for immunohistochemical staining method in140cases of human salivary gland tumors including plemorphic adenoma tissues of 32cases, basal cell adenoma tissues of 28 cases, mucoepidermoid carcinoma tissues of40 ,adenoid cystic carcinoma tissues of 40 cases and normal human salivary tissues of10 cases.To analyze the relationship between the expression and clinical pathologicalbehaviors,follow-up data were statistically analyzed by SPSS17.0 software packagewhich was used for Chi-square test and Fishers exact test.Results: The positive rate of CD147 expression in normal salivary glands, salivarygland benign tumors and salivary gland malignant tumors was 10.0%, 33.33% and80.0%,and MMP-9 was 10%, 30%, 72.5%,both respectively (P<0.05). CD147 andMMP-9 were located in the membrane of the duct epithelium in normal salivarygland,in the cytoplasm of tumour cells in salivary gland tumors, but CD147 was alsolocated in the cytoplasm of tumour cells in mucoepidermoid carcinoma and adenoidcystic carcinoma. No significant difference was found between groups in the distantmetastas and non- distant metastas on the positive rate of CD147 and MMP-9expression in mucoepidermoid, but significant difference was found in adenoid cysticcarcinoma. Significant difference was found between groups of neurotropism andnon-neurotropism on the positive rate of CD147 and MMP-9 expression in adenoidcystic carcinoma.The co- expression rate of CD147 and MMP-9 was 67.73%,and Kappawas 0.4112. Conclusion:1.CD147 and MMP-9 are objective marks to estimate the behaviors of humansalivary gland tumors,by the expression of and location of CD147 and MMP-9.2.The invading behavior of mucoepidermoid carcinoma is not related to theexpression and location of CD147 and MMP-9.3.The expression and location of CD147 and MMP-9 in the adenoid cysticcarcinoma correlates with both the nerve invasion and the distant metastasis.4.There is a signifieant positive correlation between the expression and location ofCD147 and MMP-9 ,suggesting that there will be a rational option to treat saliary glandtumors by combinating CD147 and MMP-9,especially saliary malignant tumors.
