Venenum Bufonis is prepared from the dried white secretion of the auricular and skin glands of Chinese toads (Bufo melanostictus Schneider or Bufo bufo gargarzinas Gantor). Its chemical constitution is very complex. According to domestic and foreign research reports, Venenum Bufonis can be used to treat a number of diseases, including sore throat, edema, heart failure, skin problems, and cancers. It has a wide range of clinical applications. As a natural product, the effective extracts of Venenum Bufonis has more advantages than the synthetic drugs. Double effect of Venenum Bufonis in immunomodulation and antitumor activity, and other pharmacological effects decide that it has much potential and need us to develop it. Venenum Bufonis and its agents will has broad prospects in clinical development.Our lab successfully isolated a monomer from Venenum Bufonis, named cinobufagin (CBG). In vitro, our study shows that the inhibitory effects of CBG on the proliferation of MGC-803 cells were observed in a dose-dependment and time-dependment manner. MGC-803 cells treated with CBG showed typical characteristics of apoptosis:the cells became round, cell detachment, membrane blebbing, and cell shrinkage with a condensed cytoplasm. CBG could induce G2/M phase growth arrest of MGC-803 cells in a dose-dependent and time-dependent manner and could induce apoptosis of MGC-803 cells. Meanwhile, We found that CBG treatment combined with ConA elevated the levels of Thl cytokines (TNF-α,IFN-γ). In contrast, the levels of Th2 cytokines(IL-4,IL-10) were decreased after CBG treatment combined with ConA. Taken together, these results indicated that CBG elevated Thl/Th2 cytokine production to display immunomodulation effect in vitro.Cancer is an example where the normal mechanisms of cell cycle regulation are dysfunctional, with either an overproliferation of cells and/or decreased removal of cells. In fact, suppression of apoptosis during carcinogenesis is thought to play a central role in the development and progression of some cancers. CBG has significant anti-tumor activity in vitro, we conclude that CBG can induce MGC-803 cells apoptosis through G2/M phase growth arrest to display anti-tumor activity. Meanwhile. CBG could elevated Thl/Th2 cytokine production to display immune regulation effect. The result suggested that this compound could be developed as a novel immunotherapeutic or antitumor agent to treat cancer.Chinese traditional medicines gradients are very complex. There is a lot of questions need us to resolve about the immune immunomodulation and antitumor effect of Venenum Bufonis. In other hand, Venenum Bufonis has a lot of functions, its mechanism between different organic is very complex. To demonstrate the double effect of Venenum Bufonis in immune immunomodulation and antitumor effect, and other pharmacological effects require us to do a lot of work.. The tendency of pharmacology process and the utilization of the biochemistry technology both reveal the fact that we could get the useful things from the study on Venenum Bufonis and make great contribution before long.
