Background and objective:Chronic soft tissue lesion is a kind of common disease. In current clinical therapy, silver needle therapy has been achieved a satisfactory result so far. But heating pattern of silver needle is to the needle with moxibustion or probes heating, the temperature of the needle is not easy to be controlled. To solve the problem, inner heating needle has been developed successfully in Nanjing University of Technology and Engineering. The whole needle body is evenly heated by resistance wire inside as a whole, therefore the temperature of the needle is easier to be controlled. At the same time, the mechanism for the use of silver needle therapy still rests on the clinical experiences and basic research is limited. To further explore the therapeutic mechanism of silver needle and inner heating needle therapy, we set-up the animal model of chronic soft tissue lesion in rats.The free radical metabolism, angiogenesis and the expression of bFGF are tested to analyze and compare the effects of inner heating needle and silver needle.Methods:The hitting installation is used to make dull injury on two rear limb gastroenemius of Wistar rats after half a month as a model of chronic soft tissue lesion. Sixty male Wistar rats were randomly divided into inner heating needle group (n=18), silver needle group(n=18),lesion control group(n=18) and normal control group(n=6).All of the experimental rats were executed at ld,7d and 14d after therapy. The muscle tissue of rats in different groups are observed by naked eyes and HE stain by microscope. The volume of SOD and MDA were detected, and the bFGF expression change and the number of angiogenesis in the lesion tissue was investigated by immunohistochemistry and analyzed quantifily at 1d,7d and 14d. Results:1 The change of SOD and MDA:SOD activity in inner heating needle group and silver needle group was decreased compared with that in lesion control group at 1d after treatment(P<0.05) and the value of MDA was increased (P<0.05). At 7d and 14d, Compared with lesion control group, SOD activity in inner heating needle group and silver needle group was obviously increased (P<0.01) and the value of MDA was obviously decreased (P<0.01).Furthermore, SOD activity in the former was higher than that in the latter at 7d and 14d (P<0.05) and the value of MDA was lower (P<0.05).2 The expression of bFGF:The expression of bFGF in inner heating needle group and silver needle group on Id,7d and 14d all were significantly increased than that of lesion control group (P<0.01),and the trend was increased at first and then decreased. The expression of bFGF in inner heating needle group was increased compared with that in silver needle group (P<0.05).3 The expression change of angiogenesis:angiogenesis in inner heating needle group and silver needle group was no statistical difference compared with that in lesion control group at 1d after treatment (P>0.05). Compared with lesion control group, angiogenesis in inner heating needle group and silver needle group was significantly increased at 7d and 14d (P<0.01), Moreover, the former was better than that in the latter (P<0.05)Conclusion:1 Inner heating needle and silver needle are effective on treating on chronic soft tissue lesion.2 Inner heating needle and silver needle can increase SOD activity, decrease MDA level after chronic skeletal muscle lesion in rats and enhance the body's antioxidant capacity.3 Inner heating needle and silver needle can elevate the level of the expression of bFGF and stimulate angiogenesis after chronic skeletal muscle lesion in rats.4 Inner heating needle is relatively better than silver needle in therapeutic efficacy.
