1. The establishment the acute ulcerative colitis in mice induced by DSSObjective:To explore the concentration and time of dextran sulfate sodium (DSS)to induce mice acute ulcerative coutis model.Methods:The C57BL/6 mice were fed with 3%,5%DSS solution to induce acute colitis,the control group were only fed with distilled water.The macroscopic and histopathological damages of colon were evaluated by assessing disease active index (DAI) and histological index (HI).Results:After DSS treatment, all mice had typical clinical and microscopic manifestation of colitis. Compared with NOR group,the DAI score and HI of 3%DSS group and 5%DSS group were significantly decreased. An increase of mortality rate was noted with increased DSS concentration.Conclusion:The clinical and histological features of DSS-induced acute colitis ressemble with UC in human. C57BL/6 mice feed with 3%DSS (MW36000-44000) solution for 5 days can establish an ideal acute UC model. Metheds:Male C57BL/6 mice were averagely randomized into three groups:NOR group (n=10), DSS group (n=10), and DSS+Ber (n=10).The histologial score of colitis were determined.Western blot analysis was employed to detect caspase-12 and caspase-3 protein expression in intestinal epithelial cells(IECs).The GRP78 mRNA expression in IECs was determined by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).Results:After DSS treatment, all mice had typical clinical and microscopic manifestation of colitis. Compared with DSS group, the colitis score (1.2±0.632 vs2.0±1.054,P<0.05) were significantly decreased in DSS+Ber group,and the treatment with Ber can significantly decreased the expression of caspase-12,caspase-3(P<0.05) and GRP78 mRNA(P<0.05).Conclusion:Berberine is effective on mice with colitis induced by dextran sulfate sodium, which may be associated with its preventive effect on endoplasmic reticulum stress.
